摘  要:目的利用同步辐射类同轴X线相位衬度成像技术对大鼠肝脏样本进行成像研究,旨在评价类同轴相衬成像技术在肝脏细微结构成像方面的意义。方法随机将6只C57BL大鼠分为两组,一组麻醉后直接开腹取肝脏标本,另一组麻醉后经肠系膜上静脉插管,注入生理盐水置换肝脏血管内血液,结扎肝脏血管及胆管后取出肝脏标本,经4%甲醛溶液固定后在上海光源X射线成像及生物医学应用光束站线BL13W1上进行成像,并与病理图片比较,随后对两组血管成像图像质量进行评分,测量管径并进行统计学分析。结果使用空间分辨率为0.37μm的电荷耦合感光元件(charge coupled device,CCD),注入生理盐水置换肝脏血管内血液并经干燥后,同步辐射类同轴技术能清晰显示大鼠肝脏的肝小叶结构,能分辨肝小叶中央静脉、肝板、肝窦间隙等显微解剖结构。结论同步辐射类同轴相衬成像技术能清晰显示大鼠肝脏血管及肝小叶微细解剖结构,对软组织脏器成像具有较大的应用价值,在肝纤维化及肝肿瘤早期血管成像研究上具有很大的潜力。Objective To evaluate the value of X-ray phase contrast imaging based on in-line holography with synchrotron radiation in the imaging of microstructure and microvasculature of hepatic lobule in rats. Methods Six C57BL rats were divided into two groups randomly,one group of rats were selected for saline infusion into the liver via the superior mesenteric vein to replace the blood followed by ligation of the hepatic arteries, veins and common bile duct, the other group of rats were left untreated. All of the rats livers underwent in-line holography imaging. The experiments were performed in the BL13W1 of the Shanghai synchrotron radiation facility, after being soaked in 4% formalin solution. We compared the liver images with the pathological pictures,graded the image quality from the two groups, then the statistical analysis was performed in regards to the measured vessel diameter, Results The results showed that the liver lobule structure could be visualized clearly by using in-line holography with synchrotron radiation with 0.37 μm spatial resolution charge coupled device (CCD), the microscopic anatomical structure of the central vein in hepatic lobule, hepatic plate and hepatic sinus could be distinguished especially after saline infusion and drying. Conclusions In-line holography imaging with synchrotron radiation has the ability to show the rat liver microvasculature, which has great application value for the soft tissue organ imaging and the potential in the study of the early liver fibrosis and liver tumor angiogenesis.
