摘  要:目的:研究穿心莲内酯(andrographolide,AG)对白念珠菌群感效应(quorum sensing,QS)及相关毒力基因的影响。方法:气相色谱-质谱联用仪(GC-MS)检测AG干预白念珠菌不同时间后法尼醇和酪醇的含量变化;qRT-PCR检测QS调节的相关毒力基因CHK1,PBS2,HOG1的表达。结果:白念珠菌生长2 h时法尼醇和酪醇分泌量较少,AG作用后变化不明显;12 h后分泌量最高,24 h回落。不同浓度AG干预后,法尼醇含量降低,酪醇升高,呈现剂量依赖性,其中1 000 mg·L-1AG效果均最明显。qRT-PCR结果显示,1 000 mg·L-1AG在2,12,24 h时分别使CHK1下调2.375,3.330,4.043倍;PBS2下调2.010,4.210,4.760倍;HOG1变化不明显。结论:AG可抑制法尼醇的分泌及促进酪醇的分泌,下调QS相关的毒力基因CHK1,PBS2的表达。Objective: To investigate the effect of andrographolide (AG) on quroum sensing (QS) and relevant virulence genes of Candida albicans. Method: Gas-chromatography-mass spectrometry (GC-MS) was applied to detect the changes in the content of far- nesol and tyrosol in C. albicans intervened by AG. The real-time quantitative PCR (qRT-PCR) was adopted to inspect the expressions of relevant virulence genes such as CHK1, PBS2 and HOG1 regulated by QS. Result: At 2 h after the growth of C. albican, the fame- sol and tyrosol secretions reduced, without notable change after intervention with AG. The secretions were highest at 12 h and de- creased at 24 h. After the intervention with different concentrations of AG, the farnesol content reduces, whereas tyrosol increased, in- dicating a dose-dependence, particularly with 1 000 mg · L^- 1 AG. qRT-PCR revealed that 1 000 mg · L^-1 AG could down-regulate CHK1 by 2. 375, 3. 330 and 4. 043 times and PBS2 by 2. 010, 4. 210 and 4. 760 times, with no significant change in HOG1. Conclu- sion: AG could inhibit the farnesol secretion, promote the tyrosol secretion and down-regulate QS-related virulence genes CHK1 and PBS2 expressions.
