摘 要:目的:研究丹参酮ⅡA/玉米醇溶蛋白(zein)纳米复合纤维膜的制备和力学性能、药物释放特性、透气性及抗菌性能,为该纤维膜作为伤口敷料提供实验基础。方法:以静电纺丝法制备丹参酮ⅡA/zein纳米复合纤维膜,利用扫描电子显微镜(SEM)、傅里叶红外光谱(FTIR)、X-射线衍射(XRD)对其形貌和结构进行表征;测定纺丝膜的力学性能,并考察纺丝膜的体外释放、透气性和抗菌性能。结果:当纺丝电压为20 k V,流率为0.5 m L·h-1时所纺丹参酮ⅡA/zein纳米复合纤维丝表面光滑,无药物结晶析出,药物丹参酮ⅡA以无定型状态存在,复合纤维膜中的释放为缓释,力学性能可以满足作为伤口辅料的支撑需求,膜透气性良好,对金黄色葡萄球菌和大肠杆菌抑制效果明显。结论:丹参酮ⅡA/zein复合纺丝膜可作为伤口敷料在制剂领域有潜在的应用前景。Objective: To investigate the preparation as well as the mechanical properties, drug release properties, air permeability and antibacterial properties of the tanshinone Ⅱ A/zein nanofibrous membrane for provi- ding an experimental basis for its use as a wound dressing. Methods: The tanshinone Ⅱ A/zein nanofibrous mem- brane was prepared by an electrostatic spinning method. The morphology and structure of nanofibers were character- ized by scanning electron microscope (SEM), fourier infrared (FT-IR) and X-ray diffraction (XRD). Results: In the condition of a voltage of 20 kV and a flow rate of 0.5 mL· h ^- 1, the surface of nanofibrous membrane was smooth without drug exudation, and tanshinone Ⅱ A existed as amorphous state in the nanofibrous membrane. The tanshi- none Ⅱ A/zein nanofibrous membrane showed a slow drug release, and good air permeability and mechanical prop- erties. It could effectively inhibit Staphylococcus aureus or Escherichia coll. Conclusion: The tanshinone Ⅱ A/zein nanofibrous membrane has strong potentials in development of new wound dressings.
