摘 要:目的观察金雀异黄素(genistein,GNT)对雌激素受体(estrogen receptor,ER)靶基因的转录调节作用。方法采用不同浓度的金雀异黄素(5、10、20 ng/m L)+雌二醇(10 ng/m L)或雌二醇单独处理乳腺癌细胞MCF7,36 h后收集细胞检测金雀异黄素对雌激素反应元件(estrogen response element,ERE)报告基因的转录调控作用,通过定量PCR技术检测金雀异黄素对ER靶基因PDZK1和GREB1转录水平的影响。结果随着金雀异黄素浓度的增高,ER报告基因的转录水平逐渐下降,与之相一致,PDZK1和GREB1的转录水平也逐渐下降,组间差异有统计学意义(P<0.05)。结论金雀异黄素能够与雌二醇竞争结合细胞表面的ER,并拮抗ER调控的靶基因表达。Objective To observe the transcription effect of genistein(GNT) on estrogen receptor(ER) target-gene.Methods Breast cancer cells line MCF7 were treated with GNT of different concentration(5, 10, 20 ng/m L) + estradiol(10 ng/m L) or estradiol only. Then 36 hour later, cells were collected to detect the transcription effect of GNT on report gene of estrogen response element(ERE). Quantitative PCR was applied to evaluate the expression level of ER targetgene PDZK1 and GREB1 by GNT. Results The transcription level of ER target-gene decreased gradually with the increasing of GNT concentration, and the expression level of PDZK1 and GREB1 reduced consistently with the ER transcription, the differences were statistical significance among different groups(P〈 0.05). Conclusion GNT can depress expression of ER gene by competing ER of cell surface with estradiol.
