摘  要:利用PKO无机磷培养基从不同林龄的降香黄檀-檀香根际土壤中分离筛选高效解磷菌,为进一步研制专用生物复合肥提供依据。通过溶磷圈法筛选溶磷较强(D,d值大于1.5)的菌株19株,采用钼锑钪比色法测定其解磷能力。结果显示:菌株无机磷溶量在9.23~223.27μg/mL之间,3株菌无机磷溶量在170μg/mL以上:菌株有机磷溶量在0.43~25.05μg/mL之间,有机磷溶量在15μg/mL以上有4株。采用Salkowski比色法测定菌株分泌IAA能力,10株菌具有分泌能力,其中DosaP25分泌IAA量达32.14μg/mL。综上所述,菌株DosaP15、DosaP25具有较强的解磷、分泌IAA能力。基于形态学、生理生化特性及16SrDNA序列比较分析,初步鉴定DosaPl5属于Bacillus pumilus、DosaP25属于Bacillus subtilis。试验同时表明,菌株溶磷量、分泌IAA量与培养液pH值之间不存在显著相关性。For providing the basis for further development of bio-fertilizer, high-efficiency phosphate-solubilizing bacteria were isolated and screened from rhizosphere soil of the different ages Dalbergia Odorifera and Sandalwood by PKO inorganic phosphate medium. Nineteen strains were selected based on Did values greater than 1.5, by soluble phosphorus ring method. The phosphorus solubilizing capability of the strains were measured by the molybdenum antimony resistance colorimetric method. The results showed that the inorganic phosphorus solubility of the 19 strains was between 9.23 μg/mL and 223.27 μg/mL, and among them 3 stains had the inorganic phosphate-dissolving capacity above 170 μg/mL. The organic phosphorus solubility of the 19 strains was between 0.43 μg/mL and 25.05 μg/mL, among them 4 stains had the organic phosphate-dissolving capacity above 15 μg/mL. Salkowski colorimetry measure was used to determine the IAA-producing abilities of the strains. Ten strains had the ability to produce IAA, among them the stain DosaP25 had the maximum IAA-producing capacity of 32.14 μg/mL. Consequently, the strains DosaP15, DosaP25 had the ability of efficient phosphate solubilizing and strong secretion of IAA. Based on morphology, physiological and biochemical property together with 16S rDNA sequence analysis, DosaP15 and DosaP25 was preliminarily identified as Bacillus pumilus and Bacillus subtilis, respectively. No significant correlation was observed amongst dissolved phosphorus content, IAA secretion and final pH of the culture medium.
