摘 要:目的:观察核转录因子-κB(NF-κB)、白细胞介素-17(IL-17)在胶原诱导性关节炎(CIA)模型大鼠关节滑膜细胞的表达,探讨其与炎症的关系。方法:将24只6周龄雌性Wistar大鼠随机分为空白对照组和模型对照组,每组12只。模型对照组建立CIA大鼠模型,采用足趾容积、关节炎指数(AI)评价关节炎症,酶联免疫吸附法检测IL-17,免疫印迹法检测滑膜细胞NF-κB/P65、NF-κB/P50和IκBα的表达。结果:CIA模型大鼠足趾容积大于空白对照组,模型对照组大鼠于造模后第14天开始,关节炎指数积分逐渐增加,第28天左右达到高峰。模型对照组大鼠IL-17表达高于空白对照组(P<0.01),NF-κB/P65、NF-κB/P50和IκBα蛋白活性明显高于空白对照组(P<0.05或P<0.01)。结论:CIA模型大鼠滑膜细胞NF-κB活性蛋白增高,IL-17呈高表达,可能与炎症相关。Objective:To observe the expression of NF-κB and IL-17 in joint synovial cells and serum of rats with collagen-induced arthritis and explore their relationship with inflammation.Methods:24 female Wistar rats of 6 weeks old were randomly divided into a blank control group and a model control group, 12 rats in each group.The model control group were used to establish the rat models of CIA.The toe volume and arthritis index ( AI ) were used to evaluate joint inflammation,the enzyme linked immunosorbent assay was used to detect IL-17,and the Immunoblotting was used to detect the expression of NF-κB/P65,NF-κB/P50 and IκBα in synovial cells.Results:The toe volume of the CIA model rats was larger than that of the blank control group. 14 days after the models were established,the arthritis index score of the rats in the control group gradually increased,reaching the peak by around 28 days.The expression of IL-17 in rats of the model group was higher than that of the blank group ( P 〈 0.01 ) ,and the activity of NF-κB/P65,NF-κB/P50 and IκBα protein was significantly higher than the control group ( P 〈 0.05 or P 〈 0.01 ) .Conclusion:The activity of NF-κB and the expression oflL-17 in synovial cells of CIA rat models increased,which may be associated with inflammation.
