消脂胶囊对家兔动脉粥样硬化干预的实验研究

摘　　要：[目的]观察消脂胶囊对家兔动脉粥样硬化(AS)的防治作用,为今后进一步研究提供实验依据。[方法]将实验动物分为对照组、模型组、消脂胶囊高剂量组和消脂胶囊低剂量组,于治疗结束时(给药10周后),耳缘静脉取血,检测超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)、脂联素(ADP)、C反应蛋白(CRP)和内皮素-1(ET-1)含量;同时测量血压;取心脏、主动脉、下肢动脉、肝脏,以12%甲醛溶液固定,进行组织病理学检查,对AS评分,分析得出动脉硬化指数。[结果]消脂胶囊给药10周后,消脂胶囊高剂量组家兔血清中SOD活性显著升高,消脂胶囊高、低剂量组家兔血清中ADP含量显著升高,MDA、CRP、ET-1含量明显降低,家兔收缩压下降,而消脂胶囊低剂量组家兔的舒张压下降。组织病理学结果显示,消脂胶囊高、低剂量组均能对主动脉及心肌内小动脉动脉AS及肝细胞空泡变性的病变程度有不同程度的减轻。[结论]消脂胶囊能够对家兔AS病变指标产生有效调节,调节动脉AS家兔血压,清除自由基,防止脂质过氧化,减少炎症介质产生和保护血管内皮细胞的作用,表明该药对家兔AS病变的发生和发展有较好的防治作用。[Objective] To observe the preventive effect of Xiaozhi capsule against atherosclerosis in rabbits, and provide some experi- mental evidences for further study. [Methods] Animal were divided into control group, model group high dose group of Xiaozhi capsule and and low dose group of Xiaozhi capsule. At the end of treatment （10 weeks after dosing）, ear vein blood was obtained for testing superoxide dismutase （SOD）, malondialdehyde （MDA）, nitric oxide （NO）, adiponectin （ADP）, C-reactive protein （CRP） and endothelin-1 （ET-1） con- tent; simultaneously blood pressure was measured ; the sample of heart, aorta, lower extremity arteries and liver were also harvested, fixed by 12% formaldehyde solution, and the histopathology was examined, scoring on the animal arteriosclerosis and analyzing the arterial stiffness index. [Results] Xiaozhi capsule was administered for 10 weeks. The high dose group of Xiaozhi capsule could significantly in- crease the activity of SOD in serum of rabbits. High and low dose groups of Xiaozhi capsule could significantly increase the rabbit serum level of ADP, decrease MDA, CRP and ET-1, decrease systolic blood pressure, and low dose group of Xiaozhi capsule could decrease the diastolic blood pressure. The histopathology showed that high and low dose groups of Xiaozhi capsule could alleviate lesion of atheroscle- rosis in myocardial arterioles and vacuolar degeneration of hepatocytes. [Conclusion] Xiaozhi capsule could regulate effectively the in- dex of atherosclerotic lesions in rabbit, adjust the blood pressure and scavenge free radicals, prevent lipid peroxidation, reduce inflam- mation medium and protect vascular endothelial cell. It was showed that the drug has good control effect in the occurrence and develop- ment of rabbit artery atheroselerotic lesions. The research results of the drug in the clinical prevention and treatment of atherosclerosis have provided some experimental basis.
