摘 要:目的观察中药复方制剂津力达对高脂饮食诱导的胰岛素抵抗(IR)大鼠血清瘦素、胰岛素、脂联素、抵抗素、游离脂肪酸(FFA)的影响。方法 36只大鼠随机分为正常对照组12只(正常饲料喂养)和高脂饮食组24只(高脂饲料喂养),每组随机选6只行高胰岛素-正葡萄糖钳夹试验确定成功建立IR大鼠模型,将高脂饮食组剩余18只大鼠分为高脂饮食对照组、津力达组和二甲双胍组各6只,正常对照组和高脂饮食对照组均给予生理盐水灌胃,津力达组给予津力达生理盐水混悬液灌胃,二甲双胍组给予二甲双胍生理盐水混悬液灌胃。药物干预8周后各组均行高胰岛素-正葡萄糖钳夹试验计算葡萄糖输注率(GIR),检测血清瘦素、胰岛素、脂联素、抵抗素、FFA水平。结果高脂饮食对照组GIR较正常对照组降低,津力达组及二甲双胍组GIR较高脂饮食对照组升高(P<0.05)。高脂饮食对照组较正常对照组血清瘦素、胰岛素、抵抗素及FFA升高,脂联素降低(P<0.05);津力达干预可降低血清瘦素、胰岛素、抵抗素及FFA,升高血清脂联素水平,与高脂饮食对照组比较差异有统计学意义(P<0.05)。结论津力达可以调节瘦素、胰岛素、脂联素、抵抗素等脂肪细胞因子的分泌以及血清FFA的水平,这种调节与胰岛素敏感性的改善作用有一定的关系。Objective To observe the effects of compound preparation of Chinese herbal medicine Jinlida on high-fat induced insulin resistance (IR) of serum leptin, insulin, adiponectin, resistin and free fatty acids (FFA) in rats. Methods A total of 36 rats were randomly divided into control group ( n = 12) and high-fat diet group ( n = 24). The IR models were successfully established through high-fat-diet feeding in 6 rats of the two groups randomly, which were confirmed by hyperinsulinemic euglycemic clamp test, and then were divided into high-fat control subgroup ( n = 6) , Jinlida subgroup (n = 6) and Metformin subgroup (n = 6). The control group and high-fat control subgroup were lavaged with physiological saline; Jinlida subgroup was lavaged with suspension of Jinlida and physiological saline, and Metformin subgroup was lavaged with Metformin suspension. The glucose infusion rate (GIR) was detected by hyperinsu- linemic euglycemic clamp test after the intervention in all the groups. The levels of serum leptin, insulin, adiponectin, resistin and FFA were detected. Results The GIR of high-fat control subgroup was significantly lower than that in con- trol group, while the GIRs of Jinlida and Metformin subgroups were significantly increased than that of high-fat control subgroup ( P 〈 0. 05 ). Compared with those in control group, in high-fat control subgroup the levels of serum leptin, in- sulin, resistin and FFA levels were significantly increased, while adiponectin level was significantly decreased (P 〈 0. 05). Jinlida intervention significantly reduced the levels of serum leptin, insulin, resistin and FFA, and elevated ser- um adiponectin level compared with those in hlgh-fat control subgroup ( P 〈 0.05 ). Conclusion Jinlida may regulate the secretion of adipocytokines such as leptin, insulin, adiponectin and resistin, and serum level of FFA, therefore there is some relation between the regulation and the improvement of insulin sensitivity.
