参姜锁阳益气片对寒凝气滞血瘀证大鼠脑组织中ATP酶和核苷酸的影响

摘  要:目的探讨参姜锁阳益气片对寒凝气滞血瘀证大鼠海马区与下丘脑组织中ATP酶与核苷酸的影响。方法清洁级SD大鼠60只,随机选取12只作为A组;其余48只采用肾上腺素加冰水浴复合低温冷冻法建立寒凝气滞血瘀证模型,造模成功后分为B、C、D、E组,每组12只,造模成功后第2天灌胃给药,C、D、E组分别给予参姜锁阳益气片70、140、280 mg/kg,A组和B组给予等体积的生理盐水,每天1次,连续14 d。采用分光光度法检测脑组织中Na+-K+-ATP、Ca2+-Mg2+-ATP和Ca2+-ATP酶活力,双抗体夹心法检测脑组织中环腺苷酸(c AMP)、环鸟苷酸(c GMP)、腺苷酸环化酶(AC)、磷酸二酯酶(PDE)活性。结果与A组比较,B组脑组织中Ca2+-ATP、Na+-K+-ATP、Ca2+-Mg2+-ATP酶和AC的活力显著下降,c AMP和c GMP含量减少,PDE转化率明显升高(P<0.05)。与B组比较,除C组下丘脑ATP酶活力和D组下丘脑Na+-K+-ATP酶活力及C组海马区c AMP、c GMP和下丘脑c AMP外,各组海马区及下丘脑组织ATP酶活力和c AMP、c GMP、AC均不同程度升高,PDE活性均降低,且呈剂量依赖性,以E组变化最明显(P<0.05,P<0.01)。结论参姜锁阳益气片能够提高寒凝气滞血瘀证大鼠细胞膜上多种膜转运蛋白的活性,改善c AMP和c GMP水平。Objective To study the effect of Shenjiang Suoyang Yiqi Tablets on the ATP enzyme and nucleotides in hippocampus and hypothalamus tissues of rats with cold coagulation stagnation of qi blood stasis. Methods A total of 60 SD rats were randomly divided into group A (n = 12 ) ; cold coagulation stagnation of qi blood stasis models were estab- lished in the other 48 rats by using adrenaline and icewater bath method, and then the 48 rats were randomly divided into group B, C, D and E (n = 12 for each group). On the 2nd d after models establishment, group C, D and E were given Shenjiang Suoyang Yiqi Tablets of 70, 140 and 280 mg/kg respectively with 1 time/d for consecutive 14 d, while the group A and B received an equal volume of physiological saline. Vitalities of Na ~ -K ~ -ATPase, Ca2~ -Mg~+ -ATPase and Ca2+- ATPase in brain tissues were detected using spectrophotometric method, and the activities of cAMP, eGMP, AC and PDE in brain tissues were detected using double antibody sandwich method. Results Compared with those in group A, in group B the values of Ca2+ -ATP, Na + -K + -ATP and Ca2+ -Mg2+ -ATPase of brain tissues ATP and AC activity were significantly decreased, contents of cAMP and cGMP were decreased, and PDE inversion rate was significantly increased (P 〈 0.05). Compared with those in group B, ATPase and levels of cAMP, cGMP and AC of hippoeampus and hypo- thalamus tissues in other groups were increased in different degrees, while PDE activities were decreased with dose de- pendence except for ATPase activity of hypothalamus in group C, the Na ~ -K + -ATPase activity of hypothalamus in group D and cAMP and cGMP levels of hippocampus, and cAMP level of hypothalamus in group C, while group E had the most significant change (P 〈 0. 05, P 〈 0.01 ). Conclusion Shenjiang Suoyang Yiqi Tablets may enhance the activity of a variety of membrane transport proteins in the cell membranes in rats with cold coagulation stagnation of qi blood stasis, and improve the cAMP and cGMP lev
