摘  要:目的考察不同样品溶剂对高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定黄芪甲苷的影响。方法以黄芪和感毒清颗粒为样品,以甲醇、90%甲醇、80%甲醇、70%甲醇、32%乙腈、15%乙腈和水为样品溶剂,采用HPLC-ELSD测定黄芪甲苷的含量。结果以90%甲醇溶液为样品溶剂,其系统适用性良好,准确度、精密度、线性关系等方法学验证指标符合要求。结论采用90%甲醇溶液作为样品溶剂,更有利于黄芪及制剂中黄芪甲苷的质量控制。Objective To explore the effect of sample-solvent compositions on the determination of Astragali Ⅳ by high performance liquid chromatograph- evaporative light scattering detector( HPLC-ELSD). Methods Radix astragali and ganduqing granules served as samples. Methanol,90% methanol,80% methanol,70% methanol,32% acetonitrile,15% acetonitrile,and water were sample solvents. HPLC-ELSD was used to determine content of astragaloside Ⅳ. Results The results showed that the 90% methanol solution was an appropriate sample solvent with good system suitability,precision,accuracy,and,linearity etc..Conclusion This method benefits the quality control of astragaloside Ⅳ in Radix astragali and agents containing Radix astragali.
