摘  要:基于异染原理研究聚天冬氨酸钠(PAsp Na)与2-[4-(二甲基氨基)苯乙烯]-1-乙基喹啉鎓碘化物(喹哪啶红,QR)的相互作用。利用紫外可见分光光度测定法研究QR光谱的变化,考察了离子强度、醇类和尿素等对异染效应的影响,PAsp Na与QR结合常数与热力学参数通过异染光谱的吸光度测定。结果表明QR与PAsp Na相互作用后光谱发生了剧烈变化,说明加入PAsp Na改变了溶液环境导致了异染现象。另外,外添加物如离子强度、醇类和尿素等对异染的破坏反映了异染结合物的稳定性并间接反映了二者的结合机制。PAsp Na与QR相互作用不仅可以模拟PAsp Na与药物的结合方式,还可以作为一种新的模型用QR作为探针检测生物大分子。The interaction of sodium polyaspartate( PAsp Na) and 2-[4-( dimethylamino) phenylethenyl]-1-ethyl-quinoliniuiodide( quinaldine red,QR) from the metachromasy principle was investigated. The change of QR spectrum was studied by uv-vis spectrophotometry,and the ionic strength,alcohols and urea were inspected to study the influence on metachromasy. The interaction constants and thermodynamic parameters were determined by absorbance measurements. Results showed that the dramatic change in the spectrum of PAsp Na-interacting QR indicated the presence of a completely different environment compared with that in water. The factors,such as ionic strength,alcohols and urea,on the reversal of metachromasy were discussed,and were used for determining the stability of the metachromatic complex as well as for understanding the nature of binding. QR that was strongly bound to PAsp Na could be used to model the interaction between PAsp Na and drugs,and as a new model to detect biomacromolecules.
