摘  要:目的:从胎鼠侧脑室中分离神经干细胞,并对其进行培养及鉴定。方法采用无血清培养基分离培养来自胎鼠侧脑室的神经干细胞,并诱导其分化,用免疫荧光细胞化学的方法检测BrdU、Nestin和SOX2以及神经元标记物TuJ1的表达。结果胎鼠的侧脑室能够分离培养出具有增殖能力的神经球,呈Nestin和SOX2阳性,分化诱导后产生TuJ1阳性的神经元。结论胎鼠的侧脑室能够分离培养出神经干细胞,并能够在体外增殖、诱导分化为神经元。Objective To culture and identify the neural stem cells (NSCs) in lateral ventricle of fetal mice. Methods NSCs were isolated from the lateral ventricle of fetal mice and cultured in serum-free medium. Fluorescence immunocytochemistry was carried out to examine the expression BrdU, the NSC markers Nestin and SOX2 and neuronal marker TuJ1. Results NSCs possessing proliferative capacities were obtained from the lateral ventricle of fetal mice. The cells were positive for NSC markers Nestin and SOX2, which were able to differentiate into TuJ1-positive neurons. Conclusion The cells isolated from the lateral ventricle of fetal mice are NSCs, which furthermore differentiated into neurons.
