摘  要:目的:采用星点设计-效应面法优化洋金花叶中醉茄内酯类化学成分的提取工艺。方法:以乙醇体积分数、提取时间、溶媒比为自变量,醉茄内酯总得率为因变量,通过对自变量各水平的多元线性回归及二项式拟合,采用效应面法优选提取工艺条件,同时进行预测分析。结果:确定最优提取工艺为加40倍量60%乙醇提取3次,每次提取时间70min;醉茄内酯类提取率的实测值与预测值偏差-1.56%,二项式拟合复相关系系数0.9308。结论:星点设计-效应面法优化的洋金花叶醉茄内酯提取工艺简单、精密度高,且预测性良好。Objective:To optimize the process of extracting withanolides from the leaves of Datura metel L. by central composition design/response surface methodology. Methods: With ethanol concentration, reflux time and solvent ratio as independent variables, extraction rate of withanolides from the leaves of Datura metel L. as dependent variable, levels of independent variables were regressed by multiple linear and fitted by binomial. Response surface methodology was performed to optimize extraction process and prediction analysis was carried out by comparing observed and predicted values. Results : The most suitable condition for extraction of withano- lides was as following : extracted 3 times the amount of 60% ethanol with solvent - solid ratio 40 : 1 for 70 rain at a time. The maximum yield of withanolides obtained experimentally was found to be very close to its predi- cated value with bias - 1.56% and the multiple correlation coefficiency of binomial fitting model was 0. 9308. Conclusion: This extraction process of withanolides from the leaves of Datura metel L. by central composition design/response surface methodology is a simple method with high precision and good predictability.
