摘  要:目的利用高脂高糖低蛋白饲料构建大鼠非酒精性脂肪肝模型。方法将40只SD大鼠随机分成两组,对照组20只,喂普通饲料;模型组20只,喂高脂高糖低蛋白饲料。于第5和第10周进行肝功能生化指标检查和肝脏超声检查。第10周全部处死并取肝组织作病理切片分析。结果超声声像图提示,模型组较对照组肝脏B超弥漫性光点增强,肝轮廓清。病理学检查证实模型组肝脏重度脂肪变性。两组大鼠体重变化无明显差异。模型组肝指数、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)活力明显高于对照组,除第5周谷丙氨酸转氨酶(ALT)外各项指标差异均有统计学意义(P<0.01)。结论 10周的高脂高糖低蛋白饮食可成功构建适用于脂肪肝研究的大鼠脂肪肝模型。[Objective]To establish a rat model of non-alcoholic fatty liver induced by fat-rich,high-carbohydrate and low-protein diet. [Methods]40 SD rats were randomly divided into two groups. The control group(20 cases)was given the ordinary feed,and the model group(20 cases)wad given the fat-rich,high-carbohydrate and low-protein diet. The detection of biochemical indexes about liver function and ultrasound examination was performed at 5th and 10 th week. All rats were killed and the hepatic tissues were collected for pathological analysis at 10 th week.[Results]The ultrasonographic images indicates the model group showed the enhanced diffuse light spot and clear liver contour as compared with the control group. The pathological examination confirmed that there was severe fatty degeneration of liver in the model group. There was no significant difference in weight changes between two groups. The liver index,aspartate aminotransferase(AST) activity,and alkaline phosphatase(ALP) activity in the model group were significantly higher than those in the control group,and there were significant differences in all index expect the alanine aminotransferase(ALT) activity at 5th week. [Conclusion]The fat-rich,high-carbohydrate and low-protein diet for 10 weeks can successfully induce the fatty liver rat model which is suitable for the research on fatty liver.
