摘 要:目的:了解深圳地区5岁以下腹泻患儿 A组轮状病毒的感染状况及其基因型特点。方法收集深圳市福田人民医院2007年8月~2009年2月腹泻患儿粪便标本413份,采用反转录-聚合酶链反应(RT-PCR)法检测 A组轮状病毒基因,阳性者再采用巢式 RT-PCR 分别针对病毒 VP7,VP4基因保守区进行 G/P型别检测。结果413份粪便标本 A组轮状病毒阳性率为52.5%(217/413),轮状病毒腹泻在深圳的主要流行季节是10~次年1月份,2岁以内的患儿占全部患儿总数的91.2%(198/217)。对217份轮状病毒阳性样本进行G/P分型,流行的轮状病毒G型分型依次为G3(52.1%),G1(31.3%),混合 G型感染(6.0%),G2(1.8%)及 G9(1.4%);P型分型依次为 P[8](85.7%),P[4](6.0%)及混合 P型感染(1.4%);G型和P型的组合以 G3P[8](47.5%)最为常见,其次为 G1P[8](30.4%)。结论 A组轮状病毒是深圳地区婴幼儿腹泻的主要原因,G3P[8]和 G1P[8]是2007年~2009年深圳地区的主要轮状病毒流行株,此外混合型并不少见。Objective To investigate the prevalence and genotype characteristics of group Arotavirus among children under 5 years of age with diarrhea in Shenzhen.Methods 413 fecal specimens were collected from children with acute diarrhea in Futian hospital from August 2007 to February 2009 in Shenzhen.Reverse transcription PCR(RT-PCR)were used to detect group A rotavirus.G and P genotyping were conducted to the positive specimens by using nested RT-PCR to amplify VP7 and VP4 gene.Results Group A rotavirus was detected in 217 of 413 specimens with the positive rate of 52.5%.The main epidemic season for rotavirus was from October to January in Shenzhen.The majority of children suffered rotavirus were un-der 2 years of age which account for 91.2% (198/217)of all rotavirus infections.The prevalence of G genotype was G3 (52.1%),G1 (31.3%),Gmix (6.0%),G2 (1.8%)and G9(1.4%)respectively.The prevalence of P genotype was P[8] (85.7%),P[4](6.0%)and Pmix (1.4%)respectively.G3P[8]and G1P[8]were the most common G and P genotype combinations accounting for 47.5% and 30.4% of rotavirus infections,respectively.Conclusion Group A rotavirus was the major pathogen of diarrhea among children in Shenzhen.G3P[8]and G1P[8]were the predominant genotypes from 2007 to 2009.G mix and P mix genotype were not uncommon in this region.
