摘 要:为建立大豆根系响应大豆胞囊线虫(Heterodera glycines Ichinohe,soybean cyst nematode,SCN)胁迫的差异蛋白图谱,从分子水平探索大豆抗SCN的抗病机制,本研究以抗大豆胞囊线虫3号生理小种的哈尔滨小黑豆和感病材料辽豆10号的杂交后代为材料,利用双向电泳技术,对SCN胁迫下的大豆根系进行差异蛋白质学分析。结果表明:SCN胁迫下,抗、感池材料蛋白表达发生了变化。双向电泳共检测到400余个蛋白点,抗病池材料中有3个蛋白点表现为含量上调3倍以上,9个蛋白点表现为含量下降3倍以上,抗病池中特异表达的蛋白点有6个,感病池中特异表达的蛋白点有10个。其中的16个蛋白点被鉴定,它们分别参与了植物自身的防卫反应、能量代谢、细胞信号传导和转录调控等多个生理生化过程。这些上调或下调的蛋白都是复杂的蛋白调控网络中的一部分,在植物的抗病反应中协同起着重要作用。The objective of this study was to set up a differentially expressed map for soybean root proteins induced by Heterodera glycines( SCN),and to explore the resistant mechanism of the pathogen at molecular level. A cross was made between resistant cultivar Harbin Xiaoheidou( ZDD7170) and susceptible cultivar Liaodou 10. Proteomic differentially expressed of soybean roots induced by SCN was analyzed. Results showed that,there were differences between proteins extracted from resistance pool and sensitive pool. About 400 protein spots were detected,including three 3- fold increased points and nine 3- fold decreased points in resistance pool; 6 specific proteins expressed in resistance pool and 10 specific proteins expressed in sensitive pool. 16 protein spots were obtained by searching in Mascot database and intercomparison preliminary identification. All of those proteins were probably parts of the resistant network or development related proteins which had close connection with plant defense responses,energy metabolism,cell signal transduction and transcriptional regulation.
