摘 要:目的 :探讨利用蛋白质聚丙烯酰胺凝胶电泳(PAGE)技术鉴别黄芪药材的可行性。方法 :超声提取黄芪药材的水溶性蛋白质,对其进行聚丙烯酰胺凝胶电泳,并采用凝胶成像系统分析电泳图谱。结果:黄芪药材的蛋白电泳图谱信息量丰富,不同来源黄芪药材的电泳图谱相似,存在11条明显的共有特征带,同时在条带数目、迁移率和蛋白质相对含量方面存在一定的差异性。结论:PAGE电泳具有分辨率高、谱带稳定、重现性好等优点,可用于黄芪药材的鉴别。Objectives :To explore the feasibility of using poIyacrylamide gel electrophoresis(PAGE) to identity radix astragali. Method:Water-soluble proteins in 12 batches of radix astragali were extracted by ultra-sonic and processed with PAGE,and the electrophoretogram was analyzed by using gel imaging system. Results: Electrophoretogram of radix astragali showed rich amount of information. There were obvious similarities in 11 common characteristic bands,while there were differences observed in the number of bands,relative mo- bility(Rf) and relative content of protein. Conclusion:PAGE has many advantages,such as high resolution, stable band and good reproducibility,and therefore it can be used in differentiating radix astragali.
