摘  要:目的研究热力与亚甲蓝光化学法联合应用灭活辛德毕斯病毒的效果。方法采用细胞培养法和基因检测技术,对热力与亚甲蓝光化学法联合应用灭活辛德毕斯病毒的效果进行了评价。结果经56℃水浴中热力预灭活处理1.5 h,辛德毕斯病毒滴度≤0.5 TCID50/ml,灭活对数值达到6.33 TCID50/ml;未经预灭活的辛德毕斯病毒在1μmol/L亚甲蓝+光照(MB+L)处理中随光照时间的延长,病毒残余滴度逐渐下降至检测限(TCID50/ml=0.5)。经热力灭活预处理的辛德毕斯病毒与未经预处理的病毒核酸载量之间具有相关性(R2>0.98,显著性F<0.01)。结论热力灭活预处理的辛德毕斯病毒能够反映活病毒的亚甲蓝病毒灭活效果,有望成为安全而有效的病毒灭活效果评价制品。Objective method. Methods To study the combined inactivation effect of Sindbis virus by heat and methylene blue photochemical Cell culture and gene detection technology were used to evaluate the effect of heat combined with methylene blue photochemical inactivation of Sindbis virus. Results After incubation at 56G for 1.5 h as pre - inactiva- tion, the infectivity titer of Sindbis virus was below the limit of detection ( 0. 5 TCIDs0/ml), with a decrease of 6. 33 TCID50/ml. Non - pre - inactivated virus was inactivated by 1 μmol/L methylene blue with visible light. With prolonged il- lumination time, the residual viral titers gradually decreased to the detection limit ( TCIDs0/ml = 0. 5 ). The viral load of pre - and non - pre - inactivated Sindbis virus correlated significantly ( R2 〉 0. 98, Significance F 〈 0. 01 ). Conclusion The results indicated that heat pre - inactivated Sindbis virus could reflect the effect of virus inactivation by methylene blue with visible light, which might be a safe and effective product for virus inactivation evaluation.
