‘安哥诺’李果肉中4种有机酸代谢相关酶活性测定方法的初步研究

摘　　要：以‘安哥诺’李为供试材料,采用分光光度计法,建立了‘安哥诺’李果肉有机酸代谢相关酶活性的测定方法。结果表明:测定磷酸烯醇式丙酮酸羧化酶(PEPC)活性时,需Mn2+作为激活剂,该酶的最佳透析时间为2h;测定顺式乌头酸酶(ACO)活性时,粗酶液不需要用谷胱甘肽溶液(GSH)共培养,该酶最适反应底物为柠檬酸,酶活性可达11.095U/(g·min);测定柠檬酸合成酶(CS)活性时,透析2h可测得最大酶活值为2.585U/(g·min);在酶提取液中加入0.1%的Triton X-100可使果肉中NAD-苹果酸酶(NAD-ME)活性提高38%。‘Angeleno’plum flesh were used as materials to establish methods for the determi‐nation of enzyme activities of ‘Angeleno’ plum pulp organic acid metabolism with the spectro‐photometer method . The results showed that ,Mn2＋ was needed as the activators in the process of phosphoenolpyruvate carboxylase （PEPC） activity determination ,and the optimum dialysis time of PEPC was 2 hours .It was unnecessary that the crude enzyme liquid was cul‐tured with glutathione solution （GSH） in the process of cis‐aconitase （ACO） activity determi‐nation ,The optimum reaction substrate was citric acid ,and this enzyme’s activity was up to 11 .095 U/（g·min） .The maximum activity of citrate synthase （CS）was 2 .585 U/（g·min） after the crude enzyme solution was dialysised for 2 hours .The malic enzyme （NAD‐ME） ac‐tivity increased by 38 % when 0 .1% Triton X‐100 was added to the enzyme extract .
