摘 要:目的用系统性念珠菌感染小鼠模型评价白念珠菌蛋白烯醇化酶1(Eno1)、β-葡萄糖苷酶2(Bgl2)、磷酸甘油激酶1(Pgk1)的免疫反应性。方法腹腔注射环磷酰胺致小鼠免疫力低下后,再腹腔注射白念珠菌SC5314以制备系统性念珠菌感染小鼠模型;用ELISA法和western blot法分析重组Eno1、Bgl2和Pgk1蛋白与模型小鼠血清抗体的免疫反应性。结果建模小鼠肠系膜边缘有米黄色细小颗粒,腹腔中有菌丝样生长的念珠菌,肾脏中可检测到念珠菌生长。小鼠感染白念珠菌6-8 d后出现抗Eno1、抗Bgl2和抗Pgk1的Ig G类抗体,效价峰值出现在感染后第20-24天。抗Eno1抗体效价最高,其次为抗Pgk1,抗Bgl2最低。Eno1与小鼠血清免疫反应性最强,Bgl2免疫反应性较弱。结论念珠菌可有效刺激小鼠产生抗Eno1、抗Bgl2和抗Pgk1的Ig G类抗体。重组Eno1蛋白免疫反应性最强,可用于后续研究。Objective To evaluate the immunoreactivity of Candida albicans protein enolase 1 ( Enol ), 13-glucoside 2 (Bgl2) and phos- pholycerate kinase 1 ( Pgkl ) by a systemic candiasis murine model. Methods Cyclophosphamide was used to induce immunosup- pressed murine, then intraperitoneal injection of Candida albicans SC5314 was conducted for the preparation of systemic candidiasis murine model. The immunoreactivity of Enol, Bgl2 and Pgkl with the infected mice sera was analyzed by ELISA and western blot. Re- stilts Rice yellow particles could be observed in the mesenteric marginal and Candida hyphae grow in the peritoneal cavity, Candida albicans can be detected in the infected mice kidney. In the 6th - 8th day after Candida infection, IgG antibodies of Enol, Pgkl and Bgl2 can be detected in the murine sera, and the titers reached its peak in the 20th -24th day. Enol has the highest titer, then Pgkl and then Bgl2. The three recombinant proteins show strong immunoreactivity with the infected mice sera, Enol has the highest titer and Bg12 the lowest. Conclusion Candida albicans SC5314 can effectively stimulate BALB/c mice to produce IgG antibodies against Enol, Bgl2 and Pgkl. The recombinant protein Enol show the highest immunoreactivity, and can use for follow-up study.
