摘 要:为获知升温对‘水院1号’和大连养殖刺参相关免疫酶活性及MDA含量的影响,实验设置不同温度梯度(16、20、24、28℃),分别对‘水院1号’和大连养殖刺参体腔液中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、过氧化物酶(POD)、溶菌酶(LSZ)的活性及丙二醛(MDA)含量进行测定。结果显示:(1)在升温过程中,‘水院1号’和大连养殖刺参体腔液中CAT、SOD、POD活性均呈先升高后降低的趋势。CAT活性范围分别为77.160~151.76、24.617~51.740 U/mL;SOD活性范围分别为0.070~0.154、0.056~0.100 U/mL;POD活性范围分别为2.360~3.470、1.407~2.407 U/mL。‘水院1号’和大连养殖刺参体腔液中LSZ活性均呈先升高再降低又升高的趋势,活性范围分别为9.741~30.898、8.554~28.006 U/mL。‘水院1号’刺参体腔液中MDA含量随着水温升高逐渐降低,其含量范围为42.331~46.563 nmol/mL,大连养殖群体刺参体腔液中MDA含量呈先降低后升高的趋势,变化范围为46.079~47.329 nmol/mL。(2)在升温过程中,‘水院1号’刺参体腔液中CAT、SOD、POD和LSZ活性变化较大连养殖群体刺参的变化差异大,其中CAT和POD活性变化差异显著(P〈0.05)。在28℃水温下,‘水院1号’刺参MDA含量显著小于大连养殖群体刺参MDA含量(P〈0.05)。研究表明,刺参能够对水温升高及时地做出生理免疫反应,通过调节非特异性免疫酶活性来清除机体内多余的活性氧,保护机体免受环境改变造成的损害。研究发现,‘水院1号’刺参较大连养殖群体刺参反应更为灵敏,对高温的适应性更强。本研究为探讨刺参对高温适应性和对‘水院1号’品种推广具有一定意义。The experiment studied the change of non-specific immune in rising temperature (16℃, 20℃, 24℃, 28℃). The activities of CAT, SOD, POD, LSZ and the content of MDA in Shuiyuan NO.1 sea cucumber and Dalian cultured sea were analyzed. The result show that: (1) in rising temperature,the change of CAT, SOD, POD were trend to increased and to decreased in Shuiyuan NO.1 sea cucumber and Dalian cultured sea cucumber, the activities of CAT were 77.160-151.76 U/mL and 24.617-51.740 U/mL, the activities of SOD were 0.070-0.154 U/mL and 0.056~0.100 U/mL, the activities of POD were 2.360-3.470 U/mL and 1.407-2.407 U/mL. The change of LSZ was first increased and then decreased and increased, the activities of LSZ were:9.741-30.898 U/mL and 8.554-28.006 U/mL. The content of MDA in Shuiyuan NO.1 sea cucumber was trend to increased, the change of MDA was 42.331-46.563 nmol/mL.The content of MDA in Dalian cultured sea cucumber was trend to decreased and to increased, the change was 46.079-47.329 nmol/mL.(2) In rising temperature, the change of CAT, SOD, POD, LSZ in Shuiyuan NO.1 sea cucumber were more sensitive than in Dalian cultured sea cucumber, and the change of CAT and POD had significant difference (P〈0.05). In 28℃, the content of MDA in Shuiyuan NO.1 sea cucumber was significantly less than in Dalian cultured sea cucumber (P〈0.05). The study indicated that Apostichopus japonicus (Selenka) could make certain physiological immune response to elevated temperatures, to rid the body of excess free radicals by regulating the immune defense enzyme activity, to protect body from damage caused by environmental change. Shuiyuan NO. 1 sea cucumber was more sensitive and had hight adaptation in high temperature than Dalian cultured sea cucumber. The experiment was important to study the high temperature adaptation of Apostichopus japonicus (Selenka) and to promote Shuiyuan NO.1 sea cucumber.
