摘  要:目的研究胰岛素样生长因子2(IGF2)及组蛋白乙酰基转移酶(HATs)相关因子组蛋白乙酰基转移酶(GCN5)、p300/CREB结合蛋白(p300/CBP)在正常及早期胚胎停止发育患者绒毛中的表达及三者间的相关性。方法采用免疫组化SP法检测21例正常早期妊娠绒毛、29例胚胎停止发育绒毛组织中IGF2、GCN5和p300/CBP的表达情况。结果早期胚胎停止发育绒毛中,IGF2、GCN5和p300/CBP的阳性表达显著低于正常早孕绒毛(P=0.001、P=0.002和P=0.003);并且这些指标在胚胎停止发育绒毛中的表达,两两间均存在显著正相关(r=0.563、0.622和0.563,P<0.00)。结论 IGF2、GCN5及p300/CBP的表达下调可能与早期妊娠胚胎停止发育有关,GCN5及p300/CBP的表达下调有可能参与早期绒毛发育过程中IGF2的表观遗传调控。[ Objective ] To detect the expressions of insulin-llke growth factor 2 (IGF2), histone acetyhransferases general control of nucleotidesynthesis(GCN5) and p300/CREB-binding protein(p300/CBP) in normal villus and villus of early embryoic death patients and to explore the relativity among the expressions of GCN5, p3OO/CBP and IGF2 in villus of early embryonic death. [Methods ] Immunohistochemical SP method was used to detect the expression levels of IGF2,GCN5 and p300/CBP in 21 normal early pregnancy villus samples (control group) and 29 early embryonic death villus samples. [Results] The positive expression rates of IGF2, GCN5 and p300/CBP were significantly higher in normal early pregnancy villus than those in early embryonic death villus (P =0.001, P =0.002, P =0.003). Positive correlations were found between any two of them (r =0.563/0.622/0.563, P 〈0.00). [ Conclusion ] The down-regulation of IGF2, GCN5 and p300/CBP may be related to the occurrence of early embryonic death. GCN5 and p300/CBP may take their parts in the epigenetic regulation of IGF2 in early villus development.
