摘 要:目的:研究间充质干细胞对哮喘的调控修复,观察对哮喘大鼠肺组织Notch信号通路表达的影响。方法:30只大鼠随机分为正常对照组、哮喘模型组和MSC移植组3组。造模结束后,制作肺组织石蜡切片进行病理学检查,实时荧光定量PCR(RT-PCR)方法检测Notch2和Jagged1 mRNA在大鼠肺组织中的表达水平,Western检测肺组织Notch2、Jagged1蛋白的表达。结果:哮喘组大量炎性细胞浸润,管腔狭窄,与哮喘组相比,MSC移植组气道炎症和黏液分泌显著下调,管腔黏膜无增厚,上皮细胞脱落减少,正常组无炎性改变。MSC移植组和正常组Notch2、Jagged1 mRNA表达较哮喘组下调,Notch2、Jagged1蛋白表达减少(P<0.05)。结论:间充质干细胞能影响哮喘大鼠中Notch信号通路的表达,对哮喘的治疗发挥一定的作用。Objective :To study the regulation of MSCs on the asthma;and to observe the expression of Notch signal in lung tissue of asthmatic rats. Methods : 30 rats were randomly divided into 3 group : normal control group, asthma model group and MSC transplantation group. Making paraffin sections of lung tissue for pathological examination, quantitative real-time PCR (RT-PCR) were used to identify the expression of Notch2 and Jaggedl mRNA in the rat lung tissue, and the expression of Notch2, Jaggedl were measured by Western blot. Results: Compared with normal control group and MSC transplantation group, inflammatory cell infiltration and narrower airway were observed in asthma model group . The lever of Notch2 ,Jaggedl in asthma model group was higher than MSC transplantation group and normal control group( P〈0.05 ). Conclusion: Mesenchynml stem cells can affect the expression of the Notch signaling pathway in asthmatic rats, and play a role in the treatment of asthma.
