豚鼠单侧迷路破坏后前庭内侧核中组胺受体H1亚型的变化研究

摘　　要：目的观察左侧迷路毁损后早期不同时程豚鼠前庭内侧核(MVN)组胺受体H1亚型表达特征。方法选择健康黑色豚鼠32只,按随机数字表法分为迷路破坏组(24只)和假手术组(8只),前者全部破坏左侧迷路,假手术组操作均同迷路破坏组但保持迷路完好。单侧迷路破坏术后,迷路破坏组在不同存活时间(术后12 h,48 h,5 d)随机取8只,用免疫组织化学染色,观察前庭内侧核组胺受体H1亚型的表达,并与假手术组比较。结果豚鼠经单侧迷路破坏术后,术侧MVN区组胺受体H1亚型在术后12 h、48 h较健侧及假手术组明显增加(P<0.05),但5 d时与健侧及假手术组比较,差异无统计学意义(P>0.05)。结论 (1)豚鼠MVN中存在组胺受体H1,且前庭代偿的早期极可能与MVN内组胺H1受体表达的变化有关。(2)在一侧迷路破坏后,前庭功能具有很大的代偿潜力。Objective To investigate the expression features of histamine H1 receptor in medial vestibular nucleus （MVN） of guinea pigs at different time points in the early stage after left labyrinthectomy. Methods Thirty-two healthy black guinea pigs were randomly divided into labyrinthectomy group （ n = 24） and sham-operated group （ n = 8 ）. The guinea pigs in the labyrinthectomy group received left labyrinthectomy, while those in the sham-operated group kept labyrinthine well. The expressions of histamine H1 receptor in MVH of 8 guinea pigs which were randomly selected from labyrinthectomy group 12 hours,48 hours and 5 days after unilateral labyrinthectomy were observed by immunohistochemistry,and were compared with those from sham-operated group. Results Histamine H1 receptors in MVN ipsilateral to the unilateral labyrinthectomy increased significantly 12 and 48 hours after operation compared with those in MVN contralateral to the unilateral labyrinthectomy and sham-operated group （P 〈 0. 05 ）, but showed no significant difference 5 days after operation （ P 〉 0. 05 ）. Condusion ① There is histamine H1 receptor in MVN of guinea pigs, and the early stage of vestibular compensation process may be associated with the changes in histamine H1 receptor expression in MVN.② The vestibular function has the great ability of compensation after unilateral labyrinthectomy.
