健脾补肾方对小鼠肠癌原位移植瘤肺转移及MMP-2蛋白表达的影响

摘　　要：目的:探讨健脾补肾方对肠癌原位移植瘤小鼠肺转移以及基质金属蛋白酶-2(MMP-2)蛋白表达的影响。方法:建立小鼠肠癌原位移植瘤模型,将小鼠随机分为模型组(生理盐水组)及健脾补肾方高、中、低剂量组,每组6只。各组予相应的药物每日灌胃给药1次,连续治疗14 d,于末次给药结束后处死动物,HE染色观察肺转移情况,免疫组化及Western Blot法检测MMP-2的蛋白表达。结果:健脾补肾方各剂量组的肺转移面积明显小于生理盐水组;健脾补肾方各剂量组MMP-2多呈弱或中阳性表达,而生理盐水组呈强阳性表达,MMP-2蛋白在健脾补肾方各剂量组的表达均低于生理盐水组(P<0.05)。结论:健脾补肾方具有抑制小鼠肠癌原位移植瘤模型肺转移的作用,并可能与下调MMP-2蛋白的表达有关。Objective：To investigate the effect ot~ ＂Jianpi Bushen Formula＂ （JPBSF） on lung metastasis and expression of the MMP-2 in mice. Methods： The mice models with orthotopie transplantation tumor were established and randomly divided into model group （normal saline） , JPBSF groups （high, medium and low dose） , with 6 mice in each group. Each group was treated with 1he corresponding measurements for 14 days, and all the mice were sacrificed alter the finally drug administration. Lung metastasis were observed by HE staining, and the expression of MMP-2 was absolutely detected by immunohistochemistry and Western Blot. Results： The lung metastasis area in each JPBSF group was significantly less than that of the normal saline group; expression of MMP-2 in all JPBSF groups showed mostly weak or middle positive, while in the normal saline group, it was strong positive expression. The expression of MMP-2 in each JPBSF group was lower than that of the normal saline group （ P 〈 0.05 ）. Conclusion ： JPBSF could inhibit the lung metastasis in colon cancer mice with orthotopic transplantation tumor. Its mechanism may be associated with the down-regulation of the MMP-2 protein.
