摘 要:目的:观察活血潜阳方对高血压大鼠内皮祖细胞的影响。方法:正常血压WKY大鼠10只,自发性高血压大鼠(SHR)30只(随机分为模型组、活血潜阳方组、代文组),WKY组、模型组以生理盐水2 ml/d灌胃,活血潜阳方组予活血潜阳方(0.96 g·kg-1·d-1)、代文组予代文溶液(20 mg·kg-1·d-1)灌胃,共干预8周。在实验前及灌胃4、8周时用尾动脉容积法测定血压,HE染色观察胸主动脉内皮细胞层连续性,并测定循环内皮祖细胞数量及其功能。结果:活血潜阳方组、代文组血压与模型组比较差异有统计学意义(P<0.01),代文组降压效果优于活血潜阳方组(P<0.01)。模型组大鼠胸主动脉内皮层受损最为显著,而活血潜阳方组和代文组大鼠相同部位血管内皮均获一定程度修复,且其循环内皮祖细胞数量及增殖、黏附和迁移能力显著高于模型组(P<0.01,P<0.05)。结论:SHR大鼠胸主动脉内皮细胞完整性严重破坏,循环内皮祖细胞数量减少、功能下降;活血潜阳方可提高循环内皮祖细胞数量和功能。Objective:To observe the effect of "Huoxue Qianyang Formula" on endothelial progenitor cells in rats with high blood pressure. Methods: Ten rats (WKY) with normal arterial pressure and 30 spontaneously hypertensive rats were randomly divided into model group, "Huoxue Qianyang Formula" group and Diovan group. Rats in the WKY group and model group were garaged with 2 ml normal saline per day; rats in the "Huoxue Qianyang Formula" group were treated with 0.96g·kg-1·d-1 " Huoxue Qianyang Formula" ; Diovan group was treated with 20 g·kg-1·d-1 Diovan solution, garaged for 8 weeks. Blood pressure was detected by tail artery the capacity measure before experiment, 4 weeks and 8 weeks gavaged. The continuity of continuity in thoracic aorta was observed by HE- staining. The quantity and function of endothelial progenitor cells were detected. Results: There were significant differences in the blood pressure between " Huoxue Qianyang Formula" group, Diovan group and the model group (P 〈 0.01 ), the efficacy in lower blood pressure of the Diovan group was superior to the "Huoxue Qianyang Formula" group ( P 〈 0.01 ). The damage of thoracic aorta endodermis in the model group was significant, and the blood vessel endothelium at the same area was repaired in the "Huoxue Qianyang Formula" group and Diovan group, and the quantity, proliferation, adhesiveness and transfer ability of the endothelial progenitor cells were higher than those of the model group ( P 〈 0.01, P 〈 0.05 ). Conclusion: The aortic endothelial cells integrity in thoracic aorta of SHR rats were damaged, the quantity of circulating endothelial progenitor cells were reduced and its function was declined; " Huoxue Qianyang Formula" can improve the quantity and function of circulating endothelial progenitor cells.
