紫蛇消瘤汤对子宫肌瘤模型大鼠子宫形态及PTEN蛋白表达的影响

摘　　要：目的:观察紫蛇消瘤汤对大鼠子宫肌瘤生长的抑制作用,并探讨其作用机制。方法:采用雌、孕激素联合诱导方法制备大鼠子宫肌瘤模型,将70只SD雌性大鼠随机分组,分为正常组、模型组、西药组、中成药组、中药组,各药物组于造模第21周开始给药(相应给予米非司酮片、宫瘤宁胶囊、紫蛇消瘤汤,1次/d,6次/周,连续12周)。采用光镜、电镜观察大鼠子宫组织形态学的变化,蛋白免疫印迹法(Westernblot)检测PTEN蛋白的表达。结果:病理学观察可见模型组大鼠子宫肌层多发性、局限性增生,肌细胞排列紊乱,腺体扩张;经治疗后,西药组、中成药组及中药组增生子宫肌层的改善程度明显优于模型组(P<0.05);PTEN蛋白在正常大鼠子宫组织中高表达,模型组中的表达明显减少(P<0.05),中成药组及中药组PTEN蛋白表达明显高于模型组(P<0.05)。结论:紫蛇消瘤汤可通过改善子宫基层的增生、上调大鼠子宫组织中PTEN蛋白的表达,从而抑制大鼠子宫肌瘤生长。Objective：To observe the inhibitory effect of ＂Zishe Xiaoliu Decoction＂ on uterus and PTEN expression in rats with myoma of uterus, and to discuss its mechanism. Methods： After modeling, 70 SD female rats were randomly divided into the normal group, model group, western medicine group, Chinese patent medicine group and Chinese medicine group. Each group was treated with the corresponding measurements from the 21st weeks after modeling. The histomorphological changes were observed by light microscope and electron microscope. The PTEN expression was detected by Western-blotting. Results： Pathological observation showed the muscular tissue layers increase, local muhiple of tissues, muscle ceils disarrangement, and swollen glands in the model group. After treatment, all the groups other than the control group have shown an improved lower conditions of the uterine muscle layers （ P 〈 0.05 ）. The expression of PTEN protein in uterine myoma model rats was significantly decreased （ P 〈 0.05 ）, but was increased in rats of the normal group （ P 〈 0.05 ） ; expression of PTEN protein in the Chinese patent medicine group and Chinese medicine group were higher than that of the model group （ P 〈 0.05 ）. Conclusion： ＂Zishe Xiaoliu Decoction＂ can inhibit the growth of myoma by improving the hyperplasia and up-regulating PTEN expression.
