摘  要:目的研究虎纹蛙消化道不同部位酸性磷酸酶(ACP)、碱性磷酸酶(ALP)、过氧化物酶(POX)、非特异性酯酶(NSE)、腺苷三磷酸酶(ATPase)、琥珀酸脱氢酶(SDH)等6种酶的活力分布。方法消化道分8个部位取材,应用冰冻切片、石蜡切片、酶组织化学技术及光密度定量分析。结果 ACP在十二指肠活力最高,食管、贲门和直肠其次,其余各部位酶活力显著较低(P<0.05)。ALP主要分布于空肠和十二指肠,胃体和幽门中几乎检测不出酶活力。POX在食管和胃体活力显著较高(P<0.05),在空肠和回肠酶活力显著较低(P<0.05)。NES在消化道各部位均有较多分布,其中以胃体、幽门和十二指肠酶活力显著较高(P<0.05),食管和直肠酶活力显著较低(P<0.05)。ATPase在贲门活力最高,在回肠和直肠酶活力显著较低(P<0.05)。SDH在食管、胃体和直肠活力较高,在幽门酶活力显著较低(P<0.05)。结论虎纹蛙消化道黏膜各种酶的活力分布与其他动物有一定的相似性,但其与脂类消化吸收相关酶的分布显示了明显的物种特异性。Objective To investigate the distribution of acid phosphatase(ACP),alkaline phosphatase(ALP),peroxidase(POX),non-specific esterase(NSE)adenosine triphosphatase(ATPase),and succinate dehydrogenase(SDH)in the digestive mucosa of Rana rugulosa.Methods Tissue samples were taken from eight parts of the digestive tract and studied by frozen section,paraffin section,enzyme histochemical techniques and quantitative analysis of optical density.Results The highest activity of ACP was in duodenum,followed in oesophagus,stomachus cardiacus and rectum,and the enzymatic activity was significantly lower in the rest of digestive tract(P<0.05).ALP was mainly distributed in jejunum and duodenum,but almost no enzymatic activity appeared in stomachus corpus and stomachus pyloricus.POX showed the highest enzymatic activity in oesophagus and stomachus corpus,and the lowest in jejunum and ileum(P<0.05).The enzymatic reaction of NES was stronger in the whole digestive tract,and the highest enzymatic activity was in stomachus corpus,stomachus pyloricus and duodenum,and the lowest was in oesophagus and rectum(P<0.05).ATPase showed the highest enzymatic activity in the stomachus cardiacus,but significantly lower in ileum and rectum(P<0.05).SDH showed higher enzymatic activity in oesophagus,stomachus corpus and rectum,but significantly lower in stomachus pyloricus(P<0.05).Conclusion The distribution of the digestive enzymes in the digestive mucosa of Rana rugulosa is similar to that of other animals,while the distribution of enzymes associated with lipid digestion and absorption shows obvious specificity of the species.
