摘 要:目的:研究有氧运动对中老年大鼠主动脉晚期氧化蛋白产物(Aopps)含量及主动脉管壁结构的影响,探讨规律有氧运动对主动脉的保护机制。方法:健康雌性18月龄SD大鼠30只,随机分为中老年安静组和中老年运动组,每组15只。另设6月龄年轻雌性对照组。中老年运动组进行递增负荷跑台有氧运动20周。运动结束后取胸主动脉,采用Elisa法检测胸主动脉晚期氧化蛋白产物(Aopps)含量,免疫组织化学染色检测主动脉胶原蛋白表达,透射电镜观察主动脉壁超微结构变化,应用硝酸还原酶化学比色法检测血浆一氧化氮(NO)含量,黄嘌呤氧化酶法检测血浆超氧化物歧化酶(SOD)活性,Hafeman改良法检测血浆过氧化物酶(GSH-px)活性。结果:与年轻对照组相比,中老年安静组和中老年运动组主动脉Aopps含量、胶原蛋白阳性表达均增加(P<0.05),血清中NO、SOD、GSH-px含量降低(P<0.05);与中老年安静组相比,中老年运动组主动脉Aopps含量、胶原蛋白阳性表达均降低(P<0.05),NO含量、SOD、GSH-px活性均上调(P<0.05);年轻对照组正常主动脉管腔壁结构未出现变化,中老年安静组主动脉内皮细胞水肿变形,内弹力膜厚薄不一,出现虫蚀样改变,中膜平滑肌细胞增生,胞浆内出现脂质空泡,胶原纤维增多,排列紊乱;中老年运动组大鼠主动脉内弹力膜厚薄不一但较为连续,平滑肌细胞虽出现肿胀,但较中老年安静组空泡少见。结论:通过20周的递增负荷有氧运动,中老年大鼠主动脉组织结构水平得到改善、Aopps积累减少、抗氧化酶活性提高、对氧化应激的诱导及刺激降低。运动在一定程度上延缓主动脉内皮损伤及动脉硬化进程。Objective To study the effect of 20-week aerobic exercise on the content of advanced oxidation protein product(Aopps)and structure of arterial wall in middle and old aged rats. Methods 30 healthy 18-month-old female SD rats were randomly divided into :sedentary group and exercise training group,and another 6-month-old young female rats were served as young control group. After 20-week aerobic treadmill exercise,the content of Aopps,expression of collagen and uhrastructure of arterial wall,content of NO in blood plasma,activity of SOD,and activity of GSH-px were detected and observed. Results Compared with the young control group,the positive expression of collagen in aorta appeared to be increased,and content of NO,activity of SOD and GSH-px decreased significantly in both sedentary and exercise middle and old aged groups. Compared with the sedentary middle and old aged group,the content of Aopps and the positive expression of collagen in aorta decreased significantly and the content of NO and the activity of SOD,and GSH-px increased significantly in exercise middle and old aged group. The aortic lumen wall structure in young control group remained unchanged ,while edema and deformation of the aortic endothelial cells, discrepancy of the thickness of internal elastic membrane,increased number of smooth muscle cells in tunica media vasorum,some lipid vacuoles in intracytoplasm, and increased malalinement of collagenous fiber in sedentary middle and old aged group were seen, and were lessened in exercise middle and old aged group. Conclusion Improved aortic tissue structure,decreased accumulation of Aopps,and reduced induction of oxidative stress were observed in middle and old aged people through 20-week aerobic exercise.
