摘  要:目的:探讨抗阻训练对衰老大鼠线粒体介导腓肠肌细胞凋亡通路相关因子表达的影响。方法:以雄性SD大鼠为研究对象,饲养至18月龄后通过8周抗阻训练干预,分别以无负重,30%、50%、70%最大负重进行间歇跑台运动,坡度为35°,跑速15 m/min,隔天1次。8周末检测腓肠肌ROS、Cyt c和Bcl-2 mRNA表达,并与安静组比较。结果:8周抗阻训练使衰老大鼠腓肠肌ROS、Cyt c减少,Bcl-2mRNA表达增加,其中30%、50%最大负重抗阻训练两组大鼠ROS下降具有统计学意义(P<0.01);30%、50%最大负重抗阻训练两组大鼠腓肠肌Bcl-2 mRNA表达增加具有统计学意义(P<0.01,P<0.05);无负重,30%、50%最大负重抗阻训练3组大鼠腓肠肌Cyt c减少具有统计学意义(P<0.05,P<0.01)。结论:低、中等负重抗阻训练能下调衰老过程中大鼠腓肠肌ROS、Cyt c,促进Bcl-2 mRNA的表达,起到改善线粒体功能的作用,从而预防、延迟细胞凋亡的发生。Abstract Objective To study the effects of resistance training on the factors related to mitochondria-mediated apoptosis pathway in gastrocnemius of aging rats. Methods 18-month old male SD rats underwent 8-week treadmill interval running (at the speed of 15m/min and slope of 35°)with resistance of 0%, 30% ,50% ,and 70% maximum weight,respectively. The expressions of gastrocnemius ROS,Cyt c and Bcl-2 mRNA were tested after 8-week training and compared with the sedentary control rats. Results As compared with the control group,the content of gastrocnemius ROS,Cyt c in resistance training groups decreased,especially ROS in 30% and 50% maximum weight resistance training groups (P 〈 0.01 )and Cyt c in 0%,30%,50% maximum weight resistance training groups(P 〈 0.05 ,P 〈 0.01 ), and the Bcl-2 mRNA expression in resistance training groups increased,especially in 30% and 50% maximum weight resistance training groups (P 〈 0.01 ,P 〈 0.05). Conclusion Resistance training with low and moderate intensity lowers the content of ROS and Cyt c and enhances the expression of Bcl-2 mRNA in gastrocnemius during the aging process,presenting the improvement of mitochondrial function and postponement of apoptosis occurrence.
