摘  要:目的研究丝素蛋白(SF)支架移植对大鼠脊髓损伤后胶质瘢痕形成的影响。方法64只SD大鼠随机均分为脊髓全横断术后SF支架移植组(A组)和空白对照组(B组);术后取出脊髓组织,显微镜下观察组织病理学变化,免疫组化染色和Western blot分别检测胶质纤维酸性蛋白(GFAP)、生长相关蛋白43(GAP-43)的阳性细胞染色和蛋白表达。结果与B组相比,A组可见支架内少量神经纤维,胶质细胞增生不明显,GFAP阳性细胞染色和蛋白表达减少(P<0.05),而GAP-43阳性细胞染色和蛋白表达增加(P<0.05)。结论 SF支架移植可减少大鼠脊髓损伤后胶质瘢痕形成,并可能会间接促进神经再生。Objective To investigate the effect of transplantation of silk fibroin(SF)scaffold on the formation of glial scar after spinal cord injury(SCI)in rats.Methods Sixty-four SD rats were equally randomized into two groups of A(transplantation of SF scaffold)and B(normal controls).The spinal cord tissues of rats were obtained after operation,and changes of histopathology were observed under microscope.The positive cells and protein expressions of glial fiber acid protein(GFAP)and growth-associated protein 43(GAP-43)were detected by immumohistochemical staining and Western blot,respectively.Results Compared with group B,some nerve fibers were observed in scaffold,proliferation of glial cells was not obvious,and positive cells and protein expression of GFAP were decreased(P〈0.05),while which of GAP-43 were increased in group A(P〈0.05).Conclusion Transplantation of SF scaffold can decrease the formation of glial scar after SCI in rats and may indirectly promote nerve regeneration.
