摘  要:MicroRNAs(miRNAs)是一类长度约为19-25 nt内源性的、非编码单链小RNA,在植物逆境胁迫应答中具有重要的调节作用。为了获得棉花中黄萎病抗性相关的miRNA,利用生物信息学技术,本研究从Sanger miRNA数据库和已报道的文献中找到64个与棉花黄萎病抗性相关的miRNA,预测和分析了它们的靶基因和启动子,并从中筛选出12个miRNA进行了实时荧光定量PCR分析,为进一步深入研究miRNA的作用机制奠定基础和提高转基因棉花抵抗黄萎病的能力提供新的途径。MicroRNAs (miRNAs) are a class of endogenous non-coding small RNAs in length of 19 to 25 nucleotides, which play important roles in regulation of plant stress response. To detect miRNAs related to Verticillium wilt resistance in cotton, 64 potential miRNAs were identified in cotton by mining Sanger miRNA database and the published papaers. Subsequently, the target genes and promoters of these miRNAs were predicted and analyzed by using bioinformatic approaches. Lastly, quantitative real-time PCR assays were performed to analyze the relative expression of 12 miRNAs in cotton roots. This study may pave the way for future in-depth analysis of biological action of miRNAs and provide new information for transgenic breeding for resistance to Verticillium wilt.
