摘  要:目的:探讨大黄素对软脂酸诱导的大鼠血管平滑肌细胞(VSMCs)炎症细胞因子CRP、TNF-α及i NOS的影响。方法:培养鉴定原代大鼠VSMCs,使用不同浓度的大黄素(1、10、50μmol·L^(-1))预孵育VSMCs 24 h,再以浓度为100μmol·L^(-1)的软脂酸刺激24 h,使用RT-PCR、western blot分别测定VSMCs CRP、TNF-α、i NOS的mRNA和蛋白的表达变化。结果:浓度为50μmol·L^(-1)的大黄素预孵育大鼠VSMCs 24 h后,与软脂酸组相比,CRP、TNF-α、i NOS的mRNA及蛋白的表达显著性降低,且差异有统计学意义(P<0.05)。结论:大黄素能浓度依赖性地抑制软脂酸诱导的大鼠VSMCs炎性因子的表达,可以通过抗炎而达到抗动脉粥样硬化的作用。Objective: To observe the effects of emodin on CRP, TNF-c~ and iNOS generations induced by palmitic acid in rat vascular smooth muscle cells ( VSMCs ). Methods: After VSMCs were pretreated with different concentrations of emodin( 1, 10, 50μmol·L^-1 ) for 24 h, VSMCs were treated with palmitic acid(100μmol·L^-1 ) for 24 h. At the indicated time, mRNA and protein expressions of CRP, TNF-α and iNOS in VSMCs were analyzed by RT-PCR, western blot, respectively. Results: In VSMCs, using 50 μmol·L^-1 of emodin for 24h before the stimulation with palmitic acid, mRNA and protein expressions of CRP, TNF-α and iNOS expressions were significantly decreased comparing with palmitic acid group. Conclusion: Emodin inhibits inflammatory cytokines expression induced by palmitie acid in VSMCs and has effect on atheroselerosis by its anti-inflammatory activity.
