摘 要:[目的]观察真武汤对肾阳虚型慢性肾功能衰竭(chronic renal failure,CRF)大鼠肾脏的保护作用。[方法]将30只SD大鼠随机分为正常对照组(10只)、模型组(20只),模型组给予腺嘌呤悬浊液灌胃,观察、检测大鼠肛温、抓力、肾阳虚体征症状,记录造模第21 d大鼠体重、饮水和尿量,检测大鼠尿17-OHCS含量。眼眶取血,检测大鼠血清CORT、ACTH含量,以判断模型是否成功。造模成功后将模型组大鼠分为模型组、真武汤组,真武汤组给予真武汤灌胃,连续21d。收集第21d尿量,检测其24h U-TP含量;末次给药后次日上午处死大鼠,心脏取血,检测血清BUN、Scr、AQP2含量;取肾组织HE染色,光镜下观察肾组织病理变化。[结果]腺嘌呤灌胃21 d后,大鼠体重降低,抓力减弱,肛温降低,大鼠饮水量、尿量显著增加,尿17-OHCS、血清ACTH和CORT含量显著降低,肾阳虚模型制备成功。模型组大鼠血清AQP2含量显著降低,血清Scr、BUN以及尿24h UTP含量显著升高,光镜下可见肾小球充血、间质纤维组织增生以及大量炎细胞浸润、囊腔扩大,肾小管呈代偿性扩张。真武汤组大鼠血清AQP2含量显著升高,血清Scr、BUN以及尿24h U-TP含量较模型组显著降低,肾组织病理显著改善。[结论]真武汤能够有效改善慢性肾衰竭大鼠肾功能,修复其肾组织病理损伤,改善其水液代谢紊乱。[Objective] Observe effects of Zhenwu Soup on the kidney pathology in CRF rats with kidney-yang deficiency. [Method] 30 SD rats were randomly divided into two groups: normal control group(10) and adenine model group(20). Kidney-yang deficiency model rats were made at 200 mg/kg by ig administrating adenine per day for 21 days. Body weight,rectal temperature,holding power were detected,and water intake,urine volume in 24 hours were detected and recorded. At 21 st day,the urine was collected and 17-OHCS content was assayed. And Orbital blood samples were collected to assay ACTH, CORT content. After the model was successfully made,the model group was classified into 2 groups,one treated with Zhenwu Soup. Scr,BUN,AQP2 in the serum and 24 h U-TP in the urine were detected with automatic biochemical analyzer,and kidney pathology was observed.[Results]Compared with normal control group,the model rats had lower levels of ACTH, CORT and 17-OHCS,and water metabolism was disturbed,the kidneydeficiency model was successfully made. The levels of Scr, BUN, U-TP were significantly higher. The level of AQP2 was significantly lower than the normal control group. With light microscope glomerular congestion,interstitial fibrosis and infiltration of inflammatory cells,cysts enlarged tubules showed a compensatory expansion. The levels of Scr, BUN, U-TP were lower,the level of AQP2 was higher with the treatment of Zhenwu Soup,and the pathology of glomerular and tubules improved. [Conclusion] Zhenwu Soup can effectively improve chronic renal failure rat with kidney-yang deficiency,improve the water disorder,and repair its renal pathological damage.
