中药健脾解毒汤对人大肠癌HT-29细胞的抑制作用

摘　　要：目的探讨中药健脾解毒汤基本方及其加减方对人大肠癌HT-29细胞增殖的影响。方法健脾解毒汤加减后按照功效分为基本方（基本组）、清热解毒方（清利组）、活血化瘀方（活血组）、补脾益气方（温补组）、补血益气方（滋阴组）、扶助正气方（补气血组）。各方剂水提物与人大肠癌HT-29细胞进行体外培养，光镜观察肿瘤细胞增殖变化，并用四甲基氮唑蓝比色法（MTT法）检测各组细胞活力。结果经过24h培养，各中药组细胞总体生长状态差，部分细胞形态异常，有细胞破碎；48h后，各中药组细胞稀疏，破碎明显，可见细胞死亡。MTT结果显示，各组中药对HT-29细胞均有不同程度的抑制作用，相同方剂组高浓度抑制作用强于低浓度，滋阴组在1000mg／L浓度时抑制率达到峰值（46．10％），与基本组及清利组比较差异有统计学意义（P〈0．05）。结论健脾解毒汤补血益气方具有较强的抑制大肠癌HT-29细胞增殖的作用。合理配伍是增强中药复方抗肿瘤作用的关键因素。Objective To study the effect of traditional Chinese medicinal formula basic Jianpi Jiedu decoction and its changed recipes on the proliferation of human colorectal cancer cell line HT - 29. Methods By adding different tra- ditional Chinese herbal medicine, Jianpi Jiedu decoction was divided into basic recipe, Qingre Jiedu, Huoxue Huayu, Bupi Yiqi, Buxue Yiqi, and Fuzhu Zhengqi recipes according to their function. Colorectal cancer HT - 29 cells were cul- tured with the water extract of each recipe in vitro. The proliferation of tumor ceils was observed by light microscopy and the cell vitality was detected by four methyl tetrazolium blue eolorimetric method （ MTT method）. Results After 24 h of culture, the overall cell growth condition in traditional Chinese medicine groups was poor, the morphological abnormalities of cell, cell rupture could be seen. After 48 h, the cells in Chinese medicine groups showed obviously sparse, crushing and visible cell death. MTI＂ results showed that each Chinese herbal medicine recipe had different degrees of inhibitory effect on HT -29 cells and high concentration was stronger than low concentration in the same prescription group. Inhibi- tion rate reached the peak at the concentration of 1 000 mg/L （46.10%） and had significant difference compared with the basic recipe and Qing Li group （ P 〈 0.05 ）. Conclusion Jianpi Jiedu decoction Buxue Yiqi recipe can strongly in- hibit the proliferation of human colorectal cancer cell line HT - 29. Reasonable compatibility is the key factor to enhance the anti - tumor effect of traditional Chinese medicine compound recipe.
