摘  要:目的探讨18氟-氟代脱氧葡萄糖(18 F-FDG)用于早期评价放疗对食管癌Eca109细胞生长抑制的价值。方法 Eca109细胞采用2Gy(A组)、4Gy(B组)、6Gy(C组)和8Gy(D组)照射48h后,采用18F-FDG细胞结合实验检测抑制率,CCK-8法检测细胞体外生长情况,流式细胞术检测细胞凋亡率。E组为空白对照组。结果细胞结合抑制率、细胞生长抑制率与放射剂量呈正相关(r=0.964、0.759,P<0.01)。细胞生长抑制率、细胞凋亡率与细胞结合抑制率呈正相关(r=0.736、0.995,P<0.01)。结论 18F-FDG细胞结合抑制率、细胞生长抑制率和细胞凋亡率均可用于早期评价放疗对食管癌Eca109细胞生长的抑制作用。Objective To study the role of 18F-fluorodeoxyglucose (18F-FDG) uptake in evaluating antiproliferative response of human esophageal carcinoma cells Eca109 to radiotherapy. Methods The Ecal09 cells were exposured to radiation in the doses of 2 Gy(group A) ,4 Gy(group B),6 Gy(group C) and 8 Gy(group D) for 48 hours. Group E was taken as blank control. The inhibition rate of cellular 18 F-FDG uptake was detected. The inhibition rate of cellular growth and apoptosis rate of Ecal09 cells in vetro were examined with CCK-8 and flow cytometry, respectively. Results The inhibition rates of cellular 18 F-FDG uptake and growth were positively correlated with the radiation doses(r=0. 964 and r=0. 759, P〈0. 01). The inhibition rate of cellular growth and apoptosis rate of Ecal09 cells were positively correlated with the inhibition rate of cellular ^18F-FDG uptake(r=0. 736 and r=0. 995,P〈0. 01). Conclusion As the inhibition rate of cellular growth and apoptosis rate,the inhibition rate of cellular 18F-FDG uptake can be taken as an indicator for early evaluating the inhibitory effect of radiotherapy on Eca109 cells.
