摘  要:目的观察维生素D受体激动剂(vitamin D receptor activators,VDRAs)对特应性皮炎(atopic dermatitis,AD)模型小鼠的治疗作用并探讨其在免疫方面的作用机制。方法将雌性BALB/c小鼠随机分为3组,即空白组、AD模型组和VDRAs治疗组,每组6只。用二硝基氯苯(DNCB)诱导小鼠特应性皮炎模型,共刺激9次。AD模型组和治疗组均需要造模。造模成功后,治疗组小鼠腹腔注射BXL-628,每2天注射1次,共注射7次;空白组和AD模型组不给予任何治疗。用测厚仪测量小鼠左耳中部厚度,肉眼直接观察小鼠腹部皮损变化。组织病理切片后显微镜下观察小鼠腹部皮肤厚度及炎细胞浸润情况。ELISA法测定各组小鼠血清中IL-4和IFN-γ水平。结果 VDRAs治疗组小鼠耳部肿胀度及腹部皮损炎症反应较模型组明显改善,血清IFN-γ水平高于模型组,血清IL-4水平低于模型组,差异均有统计学意义(P<0.01)。结论 VDRAs可促进AD小鼠IFN-γ分泌和抑制IL-4分泌,并调节Th1/Th2失衡,这可能是其对AD模型小鼠有一定的治疗作用的机制之一。Objective To study the therapeutic effect of VDRAs ( Vitamin D Receptor Activators ) on AD ( Atopic derma- titis model) in mice. Methods Eighteen female BABL/c mice were divided into 3 groups:blank group, AD group and VDRAs treatment group, six mice each group. Dinitrochlorobenzene (DNCB) -induced was used and each group was stimulated by DNCB 9 times except blank group. After that, BXL-628 was injected intraperito- neally in VDRAs treatment group every two day ,7 times totally. Central thickness of left ear of mice was meas- ured, and observed the changes of skin lesions and pathology in mice. The levels of IL-4, IFN-γ in serum were determined by ELISA method. Results After treatment the thickness of mice' ears and the inflammatory reac- tion were reduced in VDRAs treatment group than in AD group. In addition, the levels of IFN-γ in serum were higher than that in AD group and the levels of IL-4 were lower than that in AD group( P 〈 0.01 ) after treatment with VDRAs. Conclusion VDRAs could raise the level of IFN-γ, and lower the level of IL-4 in serum, to ad- just the balance of Th1/Th2.
