摘  要:目的探讨健脾养血祛风方对脾虚型湿疹小鼠模型皮损处细胞间黏附分子(ICAM-1及VCAM-1)的影响及可能的作用机制。方法取健康雄性小鼠50只,随机分为5组,即空白对照组、脾虚慢性湿疹模型组、脾虚慢性湿疹西药治疗组、脾虚慢性湿疹中药治疗高剂量组及低剂量组。中药组以健脾养血祛风方进行治疗,西药组以盐酸左西替利嗪治疗;采用免疫组化法检测小鼠耳部病变组织ICAM-1及VCAM-1的水平。结果模型组小鼠耳部病变组织ICAM-1及VCAM-1均有不同程度的阳性表达;给药后,脾虚慢性湿疹中药治疗高剂量组及西药组小鼠耳部病变组织ICAM-1和VCAM-1的表达均较模型组显著降低(P<0.01)。中药高剂量组与西药组ICAM-1及VCAM-1的水平差异无统计学意义(P>0.05)。结论健脾养血祛风方能够抑制脾虚型湿疹小鼠炎性细胞浸润,减轻炎症反应,其作用机制可能与抑制皮损处ICAM-1及VCAM-1的表达有关。Objective To explore the effect and possible therapeutic mechanism of JianPiYangXueQuFengFang on ICAM- 1 and VCAM-1 in mice with splenoasthenic eczema. Methods Fifty healthy male mice,were randomly divid- ed into 5 groups, namely normal control group, chronic splenoasthenic eczema control group, chronic splenoast- henic eczema therapeutic group of using western medicine, chronic splenoasthenic eczema therapeutic group of using high dose of Chinese herbs, chronic splenoasthenic eczema therapeutic group of using low dose of Chinese herbs. The Chinese medicine group were treated by the JianPiYangXueQuFengFang, the western medicine group were treated by levocetirizine hydrochloride. The level of ICAM-1 and VCAM-1 in skin lesion of mice ear were detected by immunohistochemical method. Results The positive expression of ICAM-1 and VCAM-1 in mice ear lesions of the model groups is in varying degrees;after administration, the expression of using high dose of Chinese herbs, low dose group and western medicine group were significantly lower than the model group(P 〈 0.01 ). High dose group of Chinese herbs and western medicine group showed no significant differ- ence (P 〉 0.05). Conclusion The JianPiYangXueQuFengFang can reduce inflammation in mice with chronic Splenoasthenic Eczema,and its mechanism may be associated with the inhibition of ICAM-1 and VCAM-1 ex- pression in skin lesions.
