摘  要:目的:燕窝是一种名贵药材,但目前市场上有很多假冒产品,因此需要建立一种简便可靠的鉴定方法。方法:通过荧光定量PCR对燕窝样品进行检测,对雨燕属、金丝燕属和其他物种细胞色素b基因的序列进行分析。结果:设计了一条特异靶向雨燕属和金丝燕属细胞色素b基因的TaqMan探针,发现此探针具有良好的特异性和灵敏度,可检测痕量的燕窝样品,对其他物种的检测结果为阴性。结论:所设计的TaqMan探针和实时荧光PCR方法可应用于燕窝的真伪鉴别,准确性高、实用性强。Objective: The bird's nest is a precious Chinese traditional herb, however there are a lot of counterfeit products in the market. So it is necessary to establish a simple and reliable method for identification of bird's nest. Methods: We compared the cytochrome b gene sequences of apus, collocalia and other animals and designed a TaqMan probe for real-time quantitative PCR. Results: The analysis of bird's nests by real-time quantitative PCR using this probe showed high specificity and sensitivity, and it can be used to assay trace samples of bird's nest. However it gave negative results for samples from other species. Conclusion: This probe and the method can be for identification of the bird's nest with high accuracy and practicality.
