胃瘤安不同组方对人胃癌SGC-7901增殖、细胞周期和凋亡的影响

摘　　要：目的:通过观察胃瘤安(全方、三棱莪术方、无三棱莪术方)小、中、大3个剂量作用于人胃癌SGC-7901细胞24、48h,探讨其对胃癌细胞的生长抑制作用及细胞周期和凋亡的影响,并对3种组方的胃瘤安进行比较。方法:用RPMI1640培养液对人胃癌细胞SGC-7901进行传代培养,采用MTT法检测胃瘤安不同组方对SGC-7901的生长抑制作用,采用流式细胞术检测胃瘤安不同组方对SGC-7901细胞凋亡及周期的影响。结果:胃瘤安3种不同组方均可抑制SGC-7901细胞增殖;3种不同组方的胃瘤安均可促进正常的SGC-7901细胞凋亡,细胞周期分析提示胃瘤安(全方)、胃瘤安(无三棱、莪术)将细胞SGC-7901阻滞于S期,胃瘤安(三棱、莪术)将SGC-7901细胞阻滞于G0/G1期,效应均呈浓度和时间依赖性。结论:胃瘤安3种不同组方均能抑制人胃癌SGC-7901细胞的增殖,可将细胞阻滞在S和G0/G1期,其中以胃瘤安(全方)、胃瘤安(无三棱、莪术)效果最为明显。AIM： To observe the effects of Weiliuan （compound recipe, sanleng and Ezhu, without Sanleng or Ezhu）low dose group, mid- dle dose group and high dose group on prolifera- tion and cell cycle of human gastric carcinoma cell （SGC-7901）24 and 48 hours. METHODS： Subculture was carried on by human gastric car- cinoma cell line SGC-7901 in RPMI1640 culture solution. MTT assay was used to examine the proliferation of SGC-7901 cells. Flow cytometry was used to detect the cell cycle of SGC-7901. We also treated SGC-7901 cells with three doses of Weiliuan for different times. RESULTS：MTT assay showed that three different prescriptions of Weiliuan inhibited the proliferation of SGC-7901 cell; cell cycle analysis indicated that Weili- uan （compound recipe） and Weiliuan （without Sanleng or Ezhu） arrested SGC-7901 at S phase, and Weiliuan （Sanleng and Ezhu） can arrest cell cycle of G_0/G_1 phase, which were on time and dose dependent manner. CONCLUSION. All three of the prescriptions inhibit proliferation of SGC-7901 cells,and arrest cell cycle of S or G_0/ G_1 phase. The effect of Weiliuan （compound rec- ipe） and Weiliuan （without Sanleng or Ezhu） are better than Weiliuan （sanleng and Ezhu）.
