摘 要:目的观察母鼠孕期和哺乳期暴露于壬基酚(NP)对子代肝脏增殖细胞核抗原(PCNA)的影响。方法试验分为2组:包括NP染毒组(NP 200 mg/kg·bw)和对照组(花生油)。母鼠受孕第6 d到仔鼠出生后21 d灌胃染毒壬基酚,60 d后剖杀仔鼠,检测肝脏PCNA的表达,并检测肝脏功能(包括血总蛋白TP、白蛋白ALB、前白蛋白PA、球蛋白GLB)。结果与对照组比较,NP暴露组60 d PCNA阳性细胞数明显增多(P<0.01),积分光密度增高(P<0.01)。经检测仔鼠的TP、ALB(P<0.05)与对照组差异有统计学意义,而PA、GLB(P=0.194、0.644)与对照组相比较差异无统计学意义。结论 NP暴露使仔鼠肝脏组织PCNA的表达增加,即处于增殖状态的细胞增多,并且可能对肝脏功能产生一定影响,提示NP对肝脏具有损伤作用。Objectives To observe the adverse effect of nonylphenol exposure during gestation and lactation on the expression of proliferating cell nuclear antigen ( PCNA ) in the liver of filial rats. Methods Pregnant rats were randomly assigned to an exposure group and a control group, and they were treated by garage with NP 200 mg/kg·day (exposure group) or groundnut oil (vehicle control) from the 6th day of gestation to the 21th day after giving birth. The expression of PCNA and liver functions (TP, ALB, PA and GLB) of filial rats were detected at 60 days of age. Results Compared with the control group, the number of PCNA positive cells in the liver of filial rats in exposure group was markedly increased (P 〈0.01 ) and the optical density was higher (P 〈 0.01 ). The values of TP and ALB in the exposure group were significantly different from the control group ( P 〈 0.05 ) and no significant difference on PA and GLB between the exposure and control group was observed (P = 0. 194 and 0. 644). Conclusions Exposure to NP during gestation and lactation might induce an increase of PCNA expression in the liver of filial rats, namely an increase of ceils with a state of proliferating, which indicated that an exposure of NP during gestation and lactation might have an adverse effect on the liver of pups.
