摘  要:采用高效液相色谱(HPLC)的方法,以Diamonsil C18色谱柱(4.6 mm250 mm, 5 μm),流动相为乙腈-体积分数为0.15%的磷酸水,梯度洗脱,流速为1.0 mLmin-1,柱温为30 ℃,检测波长365 nm的色谱条件同时测定了哥王中杨梅素、木犀草素、芹菜素和山柰酚含量.各成分之间分离度良好,杨梅素、木犀草素、芹菜素、山柰酚的质量浓度分别在0.100 8~1.008(r=0.999 2),0.484 8~4.848(r=0.999 0),1.354~13.54(r=0.999 6),0.316 8~3.168 mgL-1(r=0.999 0)内与峰面积呈良好的线性关系,平均回收率分别为98.5%(RSD=1.2%), 100.9%(RSD=1.7%), 99.7%(RSD=0.81%), 98.9%(RSD=1.6%).表明所建立的方法快速简便、具有较好的重复性和稳定性,为了哥王药材的质量控制提供参考.The HPLC method was established to simultaneously determine the contents of myricetin, luteolin, apigenin and kaempferol in Wikstroemia indica (L.) C. A. Mey.. The method was carried out on a Diamonsil C18 column (4.6 mm250 mm, 5 μm) eluted with the mobile phases of water containing 0.15% phosphoric acid and acetonitrile in gradient mode. The UV detection wavelength was 365 nm. The flow rate was 1.0 mLmin-1 and the column temperature was set at 30 ℃. All the standard compounds showed a good linearity in the range of 0.100 8-1.008 (r=0.999 2), 0.484 8-4.848 (r=0.999 0), 1.354-13.54 (r=0.999 6), 0.316 8-3.168 mgL-1(r=0.999 0) for myricetin, luteolin, apigenin and kaempferol, respectively. The average recoveries of these four flavonoids were 98.5%, 100.9%, 99.7% and 98.9% with RSD 1.2%, 1.7%, 0.81% and 1.6%, respectively. In conclusion, the method is simple, rapid and accurate. It can be applied for the quality control of Wikstroemia indica.keywords:Wikstroemia indica
