摘 要:目的:探究没食子酸(gallic acid,GA)对人食管癌细胞KYSE150增殖、迁移、侵袭及细胞周期与凋亡的影响。方法:用MTT、细胞划痕、transwell实验检测GA对KYSE150增殖、迁移及侵袭的影响,细胞凋亡和周期的影响,Western Blot检测细胞周期相关蛋白Cyclin D1的表达。结果:GA能抑制食管癌细胞的增殖;细胞的迁移和侵袭能力显著下降(均P<0.05);随着药物浓度增加,G0/G1期细胞比例递增,S期和G2/M期细胞比例递减,凋亡率增加(P<0.05);GA能降低细胞中Cyclin D1的表达(P<0.05)。结论:GA能抑制食管癌细胞的增殖、迁移和侵袭,能促进细胞凋亡,机制可能是通过影响Cyclin D1基因的表达使食管癌细胞阻滞于G0/G1期。Objective To explore the effects of gallic acid (GA) on the proliferation, migration, invasion, and cell cycle and apoptosis of human esophageal cancer ceil line KYSE150. Methods The effects of GA on the proliferation, migration, and invasion of KYSE150 were analyzed using MTY assay, cell scratch, and Transwell cell migration assay respectively. The apoptosis and the cell cycle of KYSE150 treated with GA were measured by flow cytometry. Expression of cell cycle-related protein CyclinD1 was detected by Western blotting. Results GA suppressed the proliferation of KYSE150. The migration and invasion of the cells were significantly decreased (P 〈 0.05 for all comparisons). With the increasing concentration of GA, the number of the cells was increased in the G0/G1 phase decreased in the S phase and G2/M phase; and the rate of apoptotic cells was increased with an elevation in the concentration of gallic acid (P 〈 0.05); the expression of CyclinD1 in KYSE150 was decreased. Conclusions Gallic acid inhibites the proliferation, migration, and invasion of KYSE150 and induces apoptosis of the cells. The mechanism of the GA-inhibited proliferation of KYSE150 may be involved in regulating the expression of CyclinD1 and arresting the cells at the G0/Gl phase.
