摘 要:以超低温保存恢复生长率较高的48h拟南芥幼苗和恢复生长率较低的72h幼苗为实验材料,研究不同苗龄幼苗在玻璃化法超低温保存装载脱水过程中内源脱落酸(ABA)、生长素(IAA)和可溶性糖组分的变化情况。结果表明,ABA在2种幼苗的装载脱水过程中含量变化很小,IAA在48h幼苗中经过装载脱水处理含量上升了近1倍。ABA和IAA原位检测发现,ABA信号在处理过程中没有明显增强,一直处于较低水平,而IAA信号在48h幼苗装载脱水过程中则显著增强,并主要分布在子叶、根部的成熟区和伸长区。2种苗龄幼苗的可溶性糖组分总含量及部分组分在装载脱水后均有所增加,且48h幼苗可溶性糖增加量大于72h幼苗。生长素对于拟南芥幼苗超低温保存装载脱水过程给予了积极的响应,可溶性糖为48h幼苗提供了较多的保护来抵御玻璃化溶液处理产生的氧化胁迫,这些都可能是导致2种苗龄幼苗恢复生长率差异的原因。48hand 72 hseedlings with high and low survival ratio after cryopreservation,respectively,were used as the experimental materials to study the changes of ABA,IAA and soluble saccharides during osmoprotection and dehydration in Arabidopsis cryopreservation.The results showed that ABA levels did not change a lot in two age seedlings cryopreservation,IAA levels increased nearly by 2-fold after osmoprotection and dehydration in 48 hseedlings.The immunohistochemistry test showed that ABA signal was at a low level without significant increase.But IAA signal was significantly increased during osmoprotection and dehydration in 48 hseedlings,and mainly distributed in cotyledons,maturation and elongation zone of roots.Total soluble saccharides and some components increased in both seedlings during the cryopreservation,and the amount of increased soluble saccharides in 48 hseedlings was more than that in 72 hseedlings.IAA showed a positive response to osmoprotection and dehydration in 48 hseedlings,and soluble saccharides provided 48 hseedlings more protection from oxidative stress,which may contribute to the difference in two age seedlings survival ratio after cryopreservation.
