摘  要:目的:观察痰瘀互结型动脉粥样硬化(AS)大鼠视网膜和肠系膜微循环的变化。方法:雄性6周龄Wistar大鼠40只,其中20只采用维生素D3(VD3)大剂量腹腔注射联合高脂乳剂长期灌胃的方法,建立痰瘀互结型AS大鼠模型(AS组),其余20只平行腹腔注射生理水以及纯净水灌胃作为对照组。对两组大鼠进行视多膜荧光造影,观察视网膜微血管形态及伴行动、静脉管径比值(DA/DV)。采用微循环观测系统分析两组大鼠肠系膜微血管形态、血流速度和DA/DV。结果:VD3大剂量腹腔注射联合高脂乳剂长期灌胃可成功建立痰瘀互结型AS大鼠模型,与对照组比较,AS组大鼠体重增加(t=5.99,P<0.05),总胆固醇水平升高(t=-13.94,P<0.05),动脉硬化指数(AI)≥4.0。AS组大鼠视网膜及肠系膜微血管数量减少、形态异常,血供不足,血液流速较对照组明显减慢(P<0.05)、微动脉变细、DA/DV明显小于对照组(P均<0.05);并可见视网膜微血管瘤及血液渗出。结论:观察痰瘀互结型AS大鼠视网膜及肠系膜微循环变化,可为临床诊疗和新药研发提供实验依据。Objective: To observe the retina and mesenteric microcirculation change of atherosclerosis (AS) rats with syndrome of intermin--gled phlegm and blood stasis. Method: There were 40 male Wistar rats, 20 of which used the method of VD3 large--dose intraperitoneal injection combined with long-term high-fat emulsion gavage , establishing the model of AS rats with syndrome of intermin--gled phlegm and blood stasis. The rest 20 of them as control group made parallel intraperitoneal injection with physiological water and pure water gavage. Contrasted success model group (AS group,n= 16)with control group and analysed retina and mesenteric microcirculation. Compared these two groups in retinal vascular morphology, arteriovenous diameter DO value(DA/DV),mesenteric mi crovessels,blood flow velocity, and the difference between DA and DV. Results: Intraperitoneal injection of large doses VD3 combined with fat emulsion long-term gavage method successfully builded the model of AS rats with syndrome of intermin--gled phlegm and blood stasis. Compared with control group, AS rats gained weight(t = 5.99, P 〈0.05), total cholesterol levels elevated(t= -13.94 ,P〈0. 05), atherosclerosis index(AI) was≥4.0 and retina and mesenteric mieroeireulation insufficiency appeared, the blood flow velocity slowed down, the number of microvessels reduced,DA/DV significantly reduced (P〈0.05) and arterioles became thinner and had blood oozing. Conclusion:The retina and mesenteric microcirculation change of the AS model rats with syndrome of intermin--gled phlegm and blood stasis can provide experimental foundation for clinical diagnosis and drug discovery.
