摘 要:目的研究多巴胺D2受体显像剂18F-Fallypride在正常食蟹猴脑内的生物分布。方法按照文献方法制备显像剂18F-Fallypride,健康雄性食蟹猴静脉注射18F-Fallypride后行动态PET图像采集,显像剂注射120 min后给予氟哌啶醇阻断试验。经图像重建后,绘制时间-活度曲线(time activity curve,TAC)。结果显像期间以双侧纹状体对称性放射性浓聚最高,其次为丘脑、海马、顶叶、枕叶,小脑的放射性浓聚最低。注药后约120 min的时间段为观察图像最佳显像期。结论 18F-Fallypride适用于活体内纹状体及纹状体外区域多巴胺D2受体的显像。18F-Fallypride放射性核素显像为多巴胺D2受体相关研究提供有力的工具。Objective To study the biological distribution of dopamine D2 receptor imaging agent18F-Fallypride in cynomolgus monkeys' brain. Methods18F-Fallypride was prepared according to the previous references. And the dynamic PET image acquisition was performed in healthy male cynomolgus monkeys after injection of 18^F-Fallypride. The blocking agent haloperidol was injected at 120 min after imaging agent injection. After image reconstruction, the time-activity curve(TAC) was created. Results The highest radioactive concentration was detected in the bilateral striatum symmetrically followed by thalamus, hippocampus, parietal lobe and occipital lobe.The lowest radioactive concentration was in the cerebellum. The optimal imaging time to observe the brain biological distribution was at120 min after18^F-Fallypride injection. Conclusions18F-Fallypride is suitable for the PET imaging in vivo striatal and extra-striatal dopamine D2 receptors. Radioactive imaging technology of18^F-Fallypride provides a powerful means for studying dopamine D2 receptors.
