利巴韦林对猫细小病毒感染诱导F81猫肾细胞中凋亡相关基因表达的影响

摘  要:试验将F81猫肾细胞随机分为对照组、药物组(猫细小病毒(FPV)+利巴韦林)、病毒组(FPV感染),采用实时荧光定量PCR的方法,检测Bcl-2、Bcl-xl、Bax和Caspase 3等凋亡相关基因的相对表达量。结果显示,与对照组相比,病毒组细胞凋亡增加,抗凋亡基因Bcl-2mRNA表达受到抑制,Bax mRNA和Caspase 3mRNA的相对表达量均有不同程度的上调。药物组细胞的抗凋亡基因Bcl-2和Bcl-xl的表达水平显著高于病毒组(P<0.05),且总体呈上调状态,但72h时Bcl-xl表达量减少;而药物组的促凋亡基因Bax和Caspase 3的表达量显著低于病毒组(P<0.05),48h前,Bax的表达量呈递减状态,72h时检测结果显示药物组Bax表达量相比对照组显著上调(P<0.05),但仍显著低于病毒组(P<0.05);与对照组相比,病毒组和药物组Caspase 3的表达量在72h时显著上调(P<0.05)。综上所述,利巴韦林可在基因水平通过上调抗凋亡基因Bcl-2和Bcl-xl和下调促凋亡基因Bax和Caspase 3的表达来抑制FPV感染诱导的F81猫肾细胞凋亡发生。The F81 cells were divided into three groups, control group, feline panleukopenia virus (FPV) infected group and ribavirin treatment group,we used Real-time fluorescent quanlitative PCR to detect the expression of Bcl-2, Bcl-xl, Bax and Caspase 3. Real-time fluorescent quanlita- tire PCR results showed that the number of apoptotic cells increased in FPV infected group com- pared with the control group, and mRNA levels of Bcl-2 was inhibited, which was an anti-apoptot- ic gene,while the expressions of Bax and Caspase 3 were both increased. In ribavirin treatment group,expression levels of Bcl-2 and Bcl-xl were significantly higher than that of FPV infected group (P〈0. 05),and had a peak at 48 h in all time points. However,mRNA levels of Bax and Caspase 3 in ribavirin treatment group were significantly lower than that of FPV infected group (P〈0.05) ,and the lowest levels were at 72 h in all time points. In a word,ribavirin could inhibit the apoptosis of F81 cells infected by FPV at the genetic level.
