槐耳清膏在体外抑制人成骨肉瘤Saos-2细胞相关实验研究

摘　　要：目的探讨槐耳清膏抑制人成骨肉瘤Saos-2的作用效果和机制。方法对数期生长人成骨肉瘤Saos-2细胞分为槐耳清膏2、8、16g/L组和空白对照组,分别采用槐耳清膏2、8、16g/L和生理盐水进行处理,分别于处理12、24、36h后,采用MTT法检测各组肿瘤细胞体外增殖情况,采用RT-PCR法检测各组肿瘤细胞趋化因子受体4(chemokine receptor 4,CXCR4)基因表达情况,采用Western blot检测各组肿瘤细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白表达水平。结果槐耳清膏16g/L组12、24、36h时细胞增殖抑制率分别为(23.30±0.10)%、(43.20±0.53)%、(51.70±0.76)%,8g/L组分别为(7.10±0.08)%、(34.90±0.33)%、(37.40±0.35)%,2g/L组分别为((1.20±0.12)%、(2.97±0.11)%、(5.30±0.17)%,空白对照组分别为(0.30±0.11)%、(1.10±0.09)%、(1.50±0.15)%),除8g/L组24h时与36h时比较差异无统计学意义外,其余各组各时间点两两比较差异均有统计学意义(P<0.05);槐耳清膏16g/L组在12、24、36h细胞CXCR4基因和VEGF蛋白表达水平明显高低于槐耳清膏2、8g/L组和空白对照组(P<0.05),槐耳清膏8g/L组低于2g/L组和空白对照组,2g/L组低于空白对照组(P<0.05);槐耳清膏2、8、16g/L组36h时CXCR4基因和VEGF蛋白表达水平低于12、24h,24h时低于12h时(P<0.05)。结论槐耳清膏在体外能抑制人成骨肉瘤Saos-2细胞的增殖、生长和转移,其作用机制可能同抑制血管生成、调节CXCR4表达有关。Objective To explore the inhibitory effect of Huaier aqueous extract on osteosarcoma Saos-2cells and its mechanism.Methods The osteogenesis sarcoma Saos-2cells in logarithmic growth phase were divided into 2,8and16g/L groups and control group,processed by 2,8and 16g/L Huaier aqueous extract and normal saline respectively.The osteosarcoma cell proliferation in vitro was detected among groups by MTT method after 12,24 and 36hours.The gene expression of chemokine receptor 4（CXCR4）was detected by RT-PCR.The protein expression of vascular endothelial growth factor（VEGF）was detected by Western blot.Results The cellular proliferation inhibition rates of12,24 and 36hours were（23.30±0.10）%,（43.20±0.53）%and（51.70±0.76）%in 16g/L group,（7.10±0.08）%,（34.90±0.33）% and（37.40±0.35）% in 8g/L group,（1.20±0.12）%,（2.97±0.11）% and（5.30±0.17）% in2g/L group,and（0.3±0.11）%,（1.1±0.09）% and（1.5±0.15）% in control group,showing no significant differences among groups and among time points（P〈0.05）except the inhibitory rate between the time points of 24 hours and 36 hours in 8g/L group（P〉0.05）.The expression levels of CXCR4 gene and VEGF protein in 12,24 and 36hours were the lowest in 16g/L group,followed by 8g/L group,2g/L group and control group in turns（P〈0.05）,and were the lowest at the time point of 36 hours,followed by 24 and 12hours in 2,8and 16g/L groups（P〈0.05）.Conclusion Huaier aqueous extract can inhibit the proliferation,growth and metastasis of osteosarcoma Saos-2cells by inhibiting angiogenesis and adjusting the CXCR4 expression.
