摘 要:目的使用mRNA探针检测Ⅱ型胶原基因型的变化,酶联免疫吸附法检测血清Ⅱ型胶原抗体浓度,阐明Ⅱ型胶原基因型改变是Legg-perthes病的病因。方法实验选用健康杂种幼犬20只(年龄为2个月,相当于人3.5岁),雌雄不限,采用自身对照方法,任选一侧髋为实验侧,另一侧作为对照侧,实验侧股骨颈内注射医用TH胶1 m L制作Legg-Perthes病动物模型,对照侧注入生理盐水1 m L。于注胶后4、8、12周时分批处死取材,采用mRNA探针检测Ⅱ型胶原基因型及酶联免疫吸附法检测血清Ⅱ型胶原抗体浓度。结果Ⅱ型胶原基因型及血清Ⅱ型胶原抗体浓度测定发现在第8~12周时实验组与对照组之间均有极显著性差异(P〈0.01)。结论Ⅱ型胶原基因型的改变可能是Legg-perthes病的病因,可能是因为Ⅱ型胶原刺激产生相应抗体发生特异性免疫反应,导致自身免疫攻击,参与Leggperthe病的发病过程。Objective To expore changes of collagen-Ⅱ genotype in Legg-perthes Disease. Methods 20 healthy halfbred young dogs were used. 1 ml of TH glue was injected into one hip joint in the experimental group,whereas 1 ml of physiological saline solution was injected into the other hip joint as control. Samples was taken at 4,8 and 12 weeks post injection.mRNA probes was used to detect the collagen genotype changes,and ELISA was used to detect the antibody thickness of collagen-Ⅱ. Results There was an notable difference of collagen-Ⅱ genotype and antibody thickness between the experimental group and the collation group at 8 and 12 weeks post injection( P〈0. 01). Conclusion The collagen-Ⅱ genotype changes may be one of the causes of Legg- perthes disease. One probable mechamism is that antibody provoked by collagen-Ⅱ lead a immunoreaction and participate in Legg- perthes morbidity process.
