摘 要:目的 观察低强度跑台运动对大鼠外周血内皮祖细胞(EPCs)数量及内皮型一氧化氮合酶(eNOS)等相关细胞因子的影响并探讨其机制.方法 8周龄SD大鼠24只,随机分为运动组与非运动组,运动组给予低强度跑台运动训练2个月,非运动组同等条件下饲养2个月.2个月后经心脏穿刺抽血,流式细胞术检测CD34+血管内皮生长因子受体(VEGFR)+ EPCs比例,酶联免疫吸附试验(ELISA)法检测血浆中eNOS、血管内皮生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)等细胞因子的浓度.结果 流式细胞术显示运动组外周血EPCs数量(0.612 ±0.018)%增加,与非运动组(0.292±0.027)%比较差异有统计学意义(P<0.01).ELISA结果显示,eNOS运动组(29.72士0.52)U/L,非运动组(22.85±1.75) U/L(P <0.01);VEGF运动组(17.05±0.34) U/L,非运动组(5.61±0.45) U/L(P<0.01);bFGF运动组(66.98±1.80) U/L,非运动组(51.21±1.90) U/L(P <0.01).以上结果显示运动组各细胞因子浓度升高,与非运动组比较差异有统计学意义.结论 低强度运动可以刺激大鼠体内细胞因子增多并增加外周血中EPCs的数量,而细胞因子的增多可能是动员SD大鼠EPCs人血和迁移的分子机制.Objective To observe the impact and explore its mechanism of low intensity treadmill exercise on the number of endothelial progenitor cells and the concentration of cytokines such as endothelial nitric oxide synthase (eNOS) in Sprague-Dawley (SD) rats.Methods Twenty four eight-weeks SD rats were randomly divided into two groups (Exercise group and Non-exercise group).Exercise group was exposed to two-month low intensity treadmill exercise,and Non-exercise group not.Both groups were harvested blood samples via cardiac puncture two months later.CD34 + vascular endothelial growth factor receptor (VEGFR) + double-positive cells (EPCs) were tested with flow cytometry,and cytokines as eNOS,vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) were detected using enzyme linked immunosorbent assay (ELISA).Results The number of EPCs increased in Exercise group (0.612±0.018) %,compared with the Non-exercise group (0.292 ±0.027) % (P〈0.01).The concentration of cytokines in Exercise group were higher than Non-exercise group as follow:eNOS (29.72 ±0.52) U/L vs.(22.85 ± 1.75) U/L (P〈0.01) ; VEGF (17.05 ±0.34) U/L vs.(5.61 ± 0.45) U/L (P〈0.01); bFGF (66.98±1.80) U/L vs.(51.21 ±1.90) U/L (P〈0.01).Conclusion Low intensity treadmill exercise increased the number of EPCs and the concentration of cytokines in SD rats.And the increment of cytokines may be the molecular mechanism to mobilize and migrate the EPCs.
